Resumo

A metilação de ilhas CpG em regiões regulatórias de vários genes tem sido descrita como um processo importante no silenciamento de genes supressores de tumor e diretamente envolvida no processo de carcinogênese de uma série de tumores. O estudo desses genes afetados pela metilação em tumores visa identificar possíveis marcadores moleculares para o diagnóstico, prognóstico e tratamento de tumores, além de possibilitar a descoberta de fatores importantes no processo biológico do câncer. Neste sentido, ao estudar o perfil diferencial de metilação de genes em carcinomas de células escamosas de cabeça e pescoço, o gene MX1 foi identificado como diferencialmente expresso. Correlação também foi encontrada entre o silenciamento de ADAM23 e estágios tardios da progressão tumoral neste tipo de câncer. Dessa forma, o presente estudo envolveu a identificação de ligantes celulares das proteínas ADAM23 e MX1 por meio de rastreamentos de duplo- híbrido, usando bibliotecas de cDNA de cérebro fetal humano. Inicialmente, foi realizado rastreamento com os domínios desintegrina e rico em cisteína (DIS-ACR) de ADAM23, com um total de 5,1x105 transformantes obtidos, dos quais somente 1 foi confirmado pelo ensaio de “plasmid linkage”. O candidato confirmado pelo “plasmid linkage”, identificado como SPRY1, foi considerado como falso positivo por ser uma proteína estritamente citoplasmática, não podendo, portanto, interagir fisicamente com os domínios DIS-ACR de ADAM23, que são extracelulares. Ainda, foi realizado rastreamento com a proteína MX1, que resultou em aproximadamente 1,0x106 transformantes, dos quais 74 foram confirmados pelo ensaio de duplo- híbrido e codificam para 9 ligantes já conhecidos e 21 ligantes prováveis de MX1. The methylation of CpG islands at regulatory regions of various genes has been described as an important process for the silencing of tumor suppressor genes and is directly involved in the carcinogenesis of a series of tumors. The study of these methylation-affected genes in tumors aims at identifying possible molecular markers for diagnostic, prognostic and treatment of such tumors, besides making possible the discovery of factors important for cancer biology. On this purpose, the study of differential pattern of gene methylation in head and neck squamous cell carcinoma identified the gene MX1 as differentially methylated. Together with this result, it was found a correlation between ADAM23 silencing and late stages of tumor progression in this type of cancer. In order to identify cellular ligands of ADAM23 and MX1, we performed two-hybrid screenings using fetal brain cDNA libraries. Initially, a screening with the disintegrin and cystein rich (DIS-ACR) domains of ADAM23 was accomplished, with an amount of 5,1x105 transformants obtained, out of which just one was confirmed by plasmid linkage assay and identified as SPRY1. However, it was considered a false positive since this protein is restricted to the cytoplasmatic compartment and, therefore, it may not physically bind the DIS-ACR domains of ADAM23, known to be extracellular. Additionally, a screening with the MX1 protein was performed, resulting in approximately 1.0x106 transformants, out of which 74 were confirmed by two-hybrid assay and revealed 9 already known and 21 putative ligands of MX1. Among the putative ligands there are proteins previously described as MX1 ligands, including MX1 itself, corroborating the results obtained in our screening.
