The species Xanthomonas citri pv. aurantifolii pathotype C (XauC), which causes type C canker, have the ability to infect only two citrus species due to the virulence variation profile and genes present in its genome, when compared to more aggressive species. There are few studies on the role of these variations in XauC, which can lead to a better characterization of the Xanthomonas-Citrus pathosystem. Thus, the identification of new genes related to the pathogenic processes of this bacterium was carried out using the screening approach of a mutant library by transposons (EZ-Tn5 System) from XauC. Two hundred and sixteen mutants were inoculated in the abaxial part of Citrus aurantifolia young leaves and the cancrose symptoms were evaluated for 25 days. Eight mutants showed altered cancer symptoms in the first inoculation in planta, however, in subsequent inoculations, seven of them returned to classic canker symptoms. The mutant, ΔXauC_P4G7, showed reproducibility in the results and revealed a total absence of type C canker classic symptoms, relating a genomic region to XauC pathogenic processes. These results contribute to a better understanding of the XauC-citrus interaction biology.
