Relações filogenéticas, filogeográficas e discriminação molecular de espécies de peixes do gênero Ancistrus Kner, 1854 (Siluriformes: Loricariidae)

Resumo

O gênero Ancistrus é um dos mais diversificados da tribo Ancistrini possuindo 64 espécies nominais. Estes peixes diferem dos outros loricariideos pela ausência de placas e odontódeos na ponta do focinho, onde existem apenas pequenos tentáculos carnudos e odontódeos interoperculares bem desenvolvidos. O gênero é caracterizado por mostrar grande variabilidade citogenética com o número diploide variando de 2n=34 e 2n=54 cromossomos. Ancistrus também se apresenta amplamente distribuído nas bacias dos rios Uruguai, Paraguai e Amazônica, sendo que as duas últimas apresentam a maior diversidade de espécies e de cariótipos do gênero. Apesar destas características, o grupo ainda é conhecido por apresentar espécies de taxonomia duvidosa e de difícil identificação, de modo que as relações filogenéticas e os padrões filogeográficos, sobretudo na bacia dos rios Paraguai e Amazonas, ainda não foram revelados. Nesse sentido, o presente trabalho teve como objetivo discriminar as diferentes linhagens de Ancistrus provenientes das bacias Amazônica e do Paraguai e utilizando a ferramenta de “DNA barcoding”, bem como identificar as relações filogenéticas e filogeográficas dessas linhagens utilizando múltiplas sequências de DNA. Foram obtidas 146 sequências do gene Citocromo Oxidase Subunidade I (COI) que foram utilizadas na discriminação das linhagens. A análise de “DNA barcoding” mostrou a ocorrência de sete linhagens de Ancistrus na bacia Amazônica e oito na bacia do Paragua. A técnica também revelou um alto valor de distância genética média entre estas linhagens (8,4%), o que indica um alto grau de isolamento genético entre as mesmas. Nas análises filogenéticas foram utilizadas 93 sequências concatenas dos genes mitocondriais ATP sintetase subunidades 6 e 8 (ATPase 6/8) e o COI e sequências do gene nuclear Rag2, enquanto na análise filogeográfica foram utilizadas apenas as sequências do gene ATPase 6/8. Na topologia gerada pelo método Bayesiano foram distinguidos 3 clados, subdivididos em 21 grupos. Os clados mostram a ocorrência de uma relação monofilética entre parte das linhagens da bacia do rio Paraguai com as linhagens da bacia amazônica, e os 21 grupos identificados apresentam altos valores de distância genética média entre si (8,4%). Na rede de haplótipos foram observados oito grandes grupos: sete pertencentes a bacia do Paraguai e um correspondendo a bacia Amazônica, não havendo compartilhamento de haplótipos entre as duas bacias. Tais resultados indicam que Ancistrus forma uma unidade monofilética nas bacias Amazônica e do rio Paraguai, sendo que estas linhagens possuem alto grau de divergência e isolamento genético entre si. Isso corrobora a existência de espécies crípticas na região e aponta a necessidade de uma revisão taxonômica para o gênero nessas bacias.
The Ancistrus genus is one of the most diverse of the Ancistrini tribe possessing 64 nominal species. These fishes differ from the other loricariids by the absence of plaques and odontodes at the tip of the muzzle, where there are only small fleshy tentacles and well-developed interopercular odontodes. The genus is characterized by showing great cytogenetic variability with the diploid number ranging from 2n = 34 and 2n = 54 chromosomes. Ancistrus is also widely distributed in the Uruguay, Paraguay and Amazon basins, with the latter two showing the greatest diversity of species and karyotypes of the genus. Despite these characteristics, the group is still known to present dubious taxonomy species that are difficult to identify, so that the phylogenetic relationships and phylogeographic patterns, especially in the Amazon and Paraguay basins, have not yet been revealed. In this sense, the present work aimed to discriminate the different Ancistrus lineages from the Amazon and Paraguay basins using the DNA barcoding tool, as well as to identify the phylogenetic and phylogeographic relationships of these lineages using multiple DNA sequences. 146 sequences of the Cytochrome Oxidase Subunit I (COI) gene were obtained, which were used to discriminate the lineages. The analysis of DNA barcoding showed the occurrence of seven lineages of Ancistrus in the Amazon basin and eight lineages in the Paraguay basin. The technique also revealed a high average genetic distance among these lineages (8.4%), indicating a high level of genetic isolation among them. In the phylogenetic analyzes, 93 concatenated sequences of the mitochondrial ATP synthase subunits 6 and 8 (ATPase 6/8) and COI and sequences of the Rag2 nuclear gene were used, whereas in the phylogeographic analysis only the ATPase 6/8 gene sequences were used. In the topology generated by the Bayesian method, 3 clades were distinguished, subdivided into 21 groups. The clades show a monophyletic relationship among the lineages from Paraguay and Amazon basins, and the 21 identified groups present high average genetic distance (8.4%). In the haplotype network, eight large groups were observed: seven belonging to the Paraguay basin and one corresponding to the Amazon basin, and there was no haplotype sharing between the two basins. These results indicate that Ancistrus forms a monophyletic unit in the Paraguay and Amazon basins, and these lineages have a high level of divergence and genetic isolation among them. This corroborates the existence of cryptic species in these regions and points out the need for a taxonomic revision for the genus in Paraguay and Amazon basins.
