Avaliação das atividades biológicas de peptídeos liberados a partir da hidrólise da caseína caprina por proteases sintetizadas por fungos filamentosos

Resumo

Peptídeos bioativos são moléculas de origem natural que podem auxiliar funções fisiológicas nos organismos. O presente estudo teve por objetivo produzir proteases por diferentes fungos filamentosos, hidrolisar a caseína caprina e avaliar o potencial de atividades biológicas dos peptídeos liberados. Baseado nos resultados de grau de hidrólise (GH), que variou entre 33,59% e 71,81% e nas atividades biológicas, os fungos Mucor subtilissimus URM 4133 e Mucor guilliermondii URM 5848 foram escolhidos para dar continuidade ao trabalho. Para a purificação de ambas as enzimas foi utilizado inicialmente um planejamento fatorial 24, com o intuito de verificar as melhores condições de produção enzimática. A atividade da protease foi máxima em pH 8,5 a 45 °C, para o M. subtilissimus URM 4133 e em pH 8,0 a 40 °C para a enzima do M. guilliermondii URM 5848; entretanto, permaneceram estáveis em praticamente toda faixa de pH e termoestáveis até 40°C e 45 °C, respectivamente. Ambas as proteases foram completamente inibidas pelo fluoreto de fenilmetanosulfonil (PMSF). As proteases purificadas de ambos os fungos foram utilizadas separadamente para hidrolisar a caseína caprina. Os hidrolisados obtidos no tempo de 12 horas apresentaram melhores resultados gerais de atividades biológicas tanto para a protease do fungo M. subtilissimus URM 4133, quanto para a protease do fungo M. guilliermondii URM 5848. A atividade antihipertensiva apresentou resultados de 92,57% e de 83,42% para a inibição da enzima conversora de angiotensina (ECA). Dentre as frações separadas em FLPC a fração P5 (URM 4133) e a fração P3 (URM 5848) apresentaram um IC50 de atividade anti-hipertensiva de 35,42 e 22,97 µg.mL-1, respectivamente. Os peptídeos liberados foram eficientes para desempenhar todas as atividades biológicas propostas, com destaque para a inibição da enzima ECA, o que mostrou o potencial destas moléculas em auxiliar no controle da pressão arterial.
Bioactive peptides are molecules of natural origin that can aid physiological functions in organisms. The present study aimed to produce proteases by different filamentous fungi, to hydrolyze caprine casein and to evaluate the potential of biological activities of the released peptides. Based on the results of hydrolysis degree (GH), which varied between 33.59% and 71.81% and in biological activities, the fungi Mucor subtilissimus URM 4133 and Mucor guilliermondii URM 5848 were chosen to continue the work. For the purification of both enzymes, a factorial design 24 was used, in order to verify the best conditions of enzymatic production. Protease activity was maximal at pH 8.5 at 45 °C for M. subtilissimus URM 4133 and at pH 8.0 at 40 °C for the M. guilliermondii URM 5848 enzyme; however, remained stable in practically every pH range and thermostable up to 40 ° C and 45 ° C, respectively. Both proteases were completely inhibited by phenylmethanesulfonyl fluoride (PMSF). Purified proteases from both fungi were used separately to hydrolyze caprine casein. The hydrolysates obtained in the time of 12 hours presented better general results of biological activities both for the protease of the fungus M. subtilissimus URM 4133 and for the protease of the fungus M. guilliermondii URM 5848. The antihypertensive activity presented results of 92.57 % and 83.42% for inhibition of angiotensin converting enzyme (ACE). Among the fractions separated in FLPC, the fraction P5 (URM 4133) and the fraction P3 (URM 5848) presented an IC50 of antihypertensive activity of 35.42 and 22.97 μg.mL-1, respectively. The peptides released were efficient to perform all the proposed biological activities, with emphasis on the inhibition of the enzyme ACE, which showed the potential of these molecules to assist in the control of blood pressure.
