Purificação de toxina killer de Aureobasidium pullulans visando o biocontrole de Penicillium digitatum e Geotrichum citri-aurantii

O setor citrícola enfrenta vários problemas com doenças que ocorrem na pós-colheita, como o bolor verde e a podridão azeda, causados por Penicillium digitatum e Geotrichum citri-aurantii, respectivamente. O surgimento de linhagens resistentes de P. digitatum e a falta de produtos registrados no Brasil para o controle da podridão azeda levam produtores e pesquisadores em busca de métodos alternativos de controle como o uso de leveduras, sendo o fator killer um dos possíveis mecanismos de ação desses microrganismos. Diante disso, esse trabalho teve por objetivo purificar a toxina killer produzida por Aureobasidium pullulans e testá-la no controle dos fitopatógenos P. digitatum e G. citri-aurantii. Inicialmente, foram testados diferentes métodos de precipitação proteica; em seguida, com a finalidade de conhecer a composição do extrato bruto proteico de A. pullulans foi determinada a atividade proteolítica e a presença de proteínas que agem sobre receptores da parede celular, β-1,3 glucanase e quitinase. A purificação da toxina foi realizada por cromatografia de exclusão molecular em coluna de gel Sephadex G-75 em tampão formiato de amônio 0,05M, pH 6,0 e cromatografia em coluna de Cellulose (Medium Fibers) em tampão citrato 0,01M, pH 4,6. Posteriormente, a purificação foi confirmada por eletroforese em gel de poliacrilamida e realizada a detecção de atividade killer em meio sólido YEPD-azul de metileno tamponado com citrato-fosfato (0,1 M pH 4,6). A identificação da toxina foi realizada por meio de espectrometria de massas (LC-MSE). Pelos resultados obtidos foi verificado que o melhor método de precipitação proteica foi etanol na proporção 2:1 (v/v etanol/sobrenadante). No estudo sobre o extrato bruto proteico da levedura foi possível observar a presença de enzimas com atividade proteolítica, atividade β-1,3-glucanase e quitinase. Durante o processo de purificação foi possível observar que a toxina killer produzida pelo isolado de A. pullulans é uma proteína de baixa massa molecular pertencente à família das ubiquitinas, que apresenta atividade killer contra os fitopatógenos P. digitatum e G. citriaurantii.
The citrus sector faces several problems with post-harvest diseases, such as green mold and sour rot, caused by Penicillium digitatum and Geotrichum citri-aurantii, respectively. The emergence of resistant P. digitatum strains and the lack of products registered in Brazil for the control of sour rot lead producers and researchers in search for alternative control methods such as the use of yeasts, being the killer factor one of the possible mechanisms of action of these microorganisms. Therefore, this work aimed at purifying the killer toxin produced by Aureobasidium pullulans and testing it in the control of P. digitatum and G. citri-aurantii phytopathogens. Initially, different methods of protein precipitation were tested; then, in order to know the composition of the protein precipitate from A. pullulans, the photeolytic activity and the presence of proteins acting on cell wall receptors, β-1,3 glucanase and chitinase were determined. Toxin purification occurred by Sephadex G-75 gel exclusion chromatography with ammonium formate elution buffer (0.05M, pH 6.0) and Cellulose chromatography (Medium Fibers) and citrate elution buffer (0.01M, pH 4.6). Subsequently, purification was confirmed by polyacrylamide gel electrophoresis and the detection of killer activity was performed in solid YEPD-methylene blue buffered with citrate-phosphate (0.1 M pH 4.6). Toxin identification was performed by mass spectrometry (LC-MSE). Based on the results obtained, it was verified that the best protein precipitation method was 2: 1 ethanol (v / v ethanol / supernatant). In the study on the yeast protein profile, it was possible to observe the presence of enzymes with proteolytic activity, β-1,3-glucanase and chitinase. During the purification process, it was possible to observe that the killer toxin produced by the A. pullulans isolate is a low molecular weight protein belonging to the ubiquitin family, presenting killer activity against P. digitatum and G. citri-aurantii phytopathogens.
