Análise citogenética comparativa de alguns representantes da família Microhylidae (Amphibia, Anura): uma abordagem clássica e molecular

Resumo

Currently, 7,414 species are described for the Anura order, with distribution practically all over the world, and 1,144 species are registered in Brazil. Today, 725 species are recognized in the Microhylidae family, divided into 12 subfamilies with more than 60 genera. Thus, through classical and molecular cytogenetic analyses, the following species were studied cytogenetically: Elachistocleis magna, Chiasmocleis shudikarensis and Dermatonotus muelleri in order to verify cytological markers that can characterize the species, prove the library's hypothesis in relation to satDNA, provide subsidies to solve taxonomic problems and add data in the literature about these animals, as there is little cytogenetic information about the group. For this study, mitotic analyzes were carried out with Giemsa for the construction of the respective karyotypes, which showed a great similarity in relation to the morphology and size of the chromosomes. It was also verified the presence and sharing in E. magna, through PCR and FISH, of three satDNA (PboSat1-176, PboSat2-173 and PboSat3-189) previously isolated from P. boiei. In this study, all satellites were amplified in the three species analyzed here. Regarding FISH of Elachistocleis magna, PboSat1-176 and PboSat2-173 did not show evident signs of labeling as a probe. PboSat3-189, on the other hand, showed signs in the chromosomes, however, in a dispersed way throughout the chromosomes, being a characteristic pattern of transposition elements. Knowing this, we can support the library hypothesis, as the species share an ancestral satDNA library, which was reduced/amplified over time. Finally, satDNAs are important to obtain data from evolutionary studies in anurans and as molecular markers, as they are present in phylogenetically distinct anurans.
Atualmente, são descritas para a ordem Anura 7.414 espécies com distribuição em praticamente todo mundo, sendo que no Brasil é registrada 1.144 espécies. São reconhecidas hoje 725 espécies na família Microhylidae, alocadas em 12 subfamílias apresentando mais de 60 gêneros. Dessa forma, por meio de análises citogenéticas clássicas e moleculares, foi estudado citogeneticamente as seguintes espécies: Elachistocleis magna, Chiasmocleis shudikarensis e Dermatonotus muellerii, com o intuito de verificar marcadores citológicos que possam caracterizar as espécies, comprovar a hipótese da biblioteca com relação aos satDNA, fornecer subsídios para resolver os problemas taxonômicos e acrescentar dado na literatura sobre esses animais, pois existem poucas informações citogenéticas a respeito do grupo. Para esse estudo foram realizadas análises mitóticas com Giemsa para a construção dos respectivos cariótipos, a qual mostrou uma grande semelhança com relação a morfologia e tamanho dos cromossomos. Também foi verificado a presença e o compartilhamento em E. magna, através de PCR e FISH, de três satDNA (PboSat1- 176, PboSat2-173 e PboSat3-189) previamente isolados de P. boiei. Neste estudo, todos os satélites foram amplificados nas três espécies aqui analisadas. Já com relação a FISH de Elachistocleis magna, PboSat1-176 e PboSat2-173 não mostraram sinais evidentes de marcação como sonda. Já PboSat3-189, mostrou sinais nos cromossomos, porém, de forma dispersa pelos cromossomos, sendo padrão característico de elementos de transposição. Sabendo disso, podemos corroborar a hipótese da biblioteca, já que as espécies compartilham uma biblioteca ancestral de satDNA, que foram reduzidas/amplificadas ao longo do tempo. Por fim, os satDNAs se mostram importantes para obter dados de estudos evolutivos em anuros e como marcadores moleculares, pois estão presentes em anuros filogeneticamente distintos.
