    Avaliação da eficácia da fibrina rica em plaquetas (FRP) em defeito crítico de calota de ratos induzidos à hipercolesterolemia

    The objective of this work was to evaluate the efficacy of platelet rich fibrin (PRF) in bone defects in rats induced hypercholesterolemia. Four experimental groups were used: a control group (CG), group PRF (GPRF), hypercholesterolemia induced clot (GCH) group and hypercholesterolemia induced PRF group, in a total of 24 animals. Hypercholesterolemia was induced by means of a special diet. To prepare the PRF in the experimental groups, it was collected in a 4ml dry tube, 1.5ml of blood, and immediately placed in a centrifuge, remaining for 10 minutes at 3000 RPM. The critical defect was performed in the calotas of the animals with a trephine of 6mm in diameter. After 28 postoperative days, euthanasia was performed for histological and microtomographic analysis, evidencing an increase in bone formation in all groups of the study.     Through a brief review of the literature the evolution of adhesive cementation during all these years was explored with the purpose of exposing the different cementation protocols of the different types of ceramic systems, thus helping the dental surgeon in the daily clinic. Based on the information contained, it was concluded that the materials used in the fabrication of indirect restorations as well as the cements and techniques used to use these materials are constantly evolving. Thus it is necessary that the professionals of the area are in continuous update of the knowledge so that it can make use, with enough responsibility, of everything that modern dentistry can offer

    Abordagem odontológica aos distúrbios do sono da criança e do adolescente: relato de caso clínico

    Obstructive Sleep Apnea and Obstructive Sleep Hypopnea Syndrome (OSAHS) is a disease that interferes with the growth and development of the child, which may have a harmful impact on the health of the pediatric patient, being an important and relevant predominance.This is a topic that is increasingly gaining importance and is considered a public health problem since it affects these patients significantly.Through several studies, the association between OSAHS and presence of craniofacial alterations and dental malocclusion, such as maxillary atresia and posterior crossbite, respectively, was shown. The present study aims to show, from the present clinical case, the significant improvement of the OSAHS in children, comparing some three-dimensional analyzes of the upper airways obtained by tomography, and the quantitative difference of apnea episodes by polysomnography, before and after the dental intervention through Rapid Maxilla Expansion (RME)

    Expectativa e prática materna do aleitamento exclusivo e a saúde bucal do bebê

    The exclusive breastfeeding (EBF) is fundamental for the health of child, however the sucess of the practice isn't always accomplished, because it depends on different determinants. In view of this, the objective of the present study was to verify the expectations of pregnant women in relation to exclusive breastfeeding and the importance of breastfeeding and its relationship with the oral health of the child and then accompany the practice during the first six months of the baby's life for making inquiries and oral examination of the mother and baby. It is a longitudinal study with 74 pairs of mothers and children (n=148). The pregnant women who were in the last trimester of gestation were interviewed about the expectation of the EBF and its relationship with the baby's oral health in the Family Health Units. At six months of age, the mothers were visited at home to verify the practice of EBF and to perform oral examination. It was found that 95.95% of respondents wanted to breastfeed her child; 74% wanted EBF up to six months old baby, however, after six months, 63.51% of mothers were breastfeeding and only 18% were on EBF; 31.05% of the mothers had trouble breastfeeding. Regarding oral health 98.65% of the women wanted to clean the baby's mouth and take him to the dentist; 36.49% lead pacifier to the hospital and 29.73% felt that breastfeeding was not important to the oral health of the baby. After six months of the baby's birth; 63.51% cleared the baby's mouth; 91.89% had not yet taken their babies to the dentist and 25.68% led pacifier to the hospital. Regarding oral health, 32.72% of the mothers had a white spot. Of the total of 74 babies, 16 had teeth at six months of age, three had white spots and were not in EBF.     Dimorfismo sexual relacionado às vias de reabsorção e formação de tecido ósseo de ratos e ratas castrados, após a exodontia do incisivo superior: avaliação microtomográfica e por PCR em tempo real

    The osteoporosis is a systemic, multifactorial and progressive disease. It is typified decrease, deterioration and modification of macro and microarchitecture of bone tissue. A long time, it was considered a woman illness, however, it is being recognized as a man illness. Thus, the objective of this study was to observe how the resorption and formation pathways behave in the animals with predisposition to osteoporosis, both male and female. For this purpose, 30 male rats and 30 female rats were used and divided into groups: SHAM (♀/♂) and castrated, that they were orchiectomized ♂ (ORQ) and ovariectomized ♀ (OVX). In this way, after 30 days of the castration, the animals were submitted to extraction of right upper incisor and divided in according with the euthanasia time, that it was 14 and 42 days after the extraction surgery and analysis method of the alveolus in repair, real time Polymerase Chain Reaction (PCR) and microcomputerized tomography (micro-ct). The quantitative data were submitted to Shapiro Wilk test, Anova test and Tukey post-test with 5% of significance level. In the gene expression analysis, it was observed that the alveolar repair process after 14 days of extraction surgery, in OVX showed a significant increase of gene expression coding for alkaline phosphatase (ALP) and osteocalcin (OC). In ORQ, results showed a peak of gene expression for ALP and receptor activator of nuclear factor kappa-β ligand (RANKL). However, after 42 days of extraction surgery, OVX and ORQ showed a decrease in gene expression for osteoprotegerin and RANKL. For micro-ct analysis of alveolus repair after 42 days extraction surgery, OVX showed low percentage of bone volume and the largest separation between bone trabecular and right percentage of bone porosity.     Efeitos da reabilitação oral com novas próteses totais convencionais sobre a força de mordida e sua correlação com a dimensão vertical de oclusão

    The aging causes a series of functional chances that occur in the whole body, including the stomatognatic system. A large portion of elderlies suffer tooth loss along life and need prosthetic treatment to recover the masticatory function. The muscle evaluation before and after rehabilitation treatment is justified to the knowledge of these individuals muscle function in different oral situations. The aim of this study was to assess the effects of oral rehabilitation with new conventional complete dentures on the bite force and its correlation with occlusal vertical dimension (OVD). Patients from Complete Dentures Clinic from Araçatuba Dental School (UNESP) were selected by anamnesis according to the pre-established inclusion and exclusion criteria. At the first moment, the patient's selection was made and demographic data were collected. After that, the bite force exams were made while the patients still wore their old prostheses (T0). 30 (T1) and 100 (T2) days after the new prostheses implantation, the exams were repeated, and the OVD measurements with both old and new prostheses were made at 30 days to comparison. The data were submitted to the 2-way ANOVA, honestly significant difference Tukey (HSD) and Pearson correlation tests, all with 5% significance. Ten patients were enrolled (9 women and 1 men) with mean are 66,1 years (range 51 to 87 years). They wore the old prostheses for mean 12,9 years (range 05 to 30 years). Although there were numeric changes of bite force values after the rehabilitation, there was no statistical difference among the analysis periods and there was positive correlation between OVD and bite force at T2. Thus, the rehabilitation did not cause statistical difference in bite force along the time, but the higher the OVD recovery the higher the bite force at T2

    Análise imunoistoquímica no tecido ósseo alveolar da influência da cisplatina na periodontite experimental em ratos

    The aim of this animal study evaluate via photometric analysis by micro-computed tomography (Micro-CT) and analysis of immunohistochemically study, the influence of chemotherapeutic drug Cisplatin (CIS) on metabolism of alveolar bone in the evolution of experimental periodontitis (EP) in rats. In the present study, 120 male rats divided following a table generated by a computer program into 4 groups, with 30 animals each. NEP- SS group, also called the sham group. The animals in this group received two intraperitoneal injections with 48-hour interval between them of 0.5 ml of saline solution 0.9 % (SS). These animals were not induced to EP. EP - SS group: the animals in this group received two intraperitoneal injections with 48-hour interval between them of 0.5 ml of SS. Soon after the first injection, PE was induced in the lower first right and left molars. NEP - CIS group: the animals in this group received two intraperitoneal injections with 48- hour interval between them of 5 mg/kg and 2.5 mg/kg of cisplatin (CIS) respectively. These animals were not induced to EP. EP - CIS group: the animals in this group received two intraperitoneal injections with 48-hour interval between them of 5 mg/kg and 2.5 mg/kg CIS respectively. Soon after the first injection, PE was induced in the lower first right and left molars. EP was induced by the placement of a number 24 cotton thread around the lower first molars. After of 07, 15 and 30 days after the first intraperitoneal injection (SS or chemotherapy) animals were euthanized using an overdose of thiopental (150mg/kg). The mandibles collected were divided into two parts and processed according each analyzes. For the photometric analysis was evaluated bone loss (EBL) in the first molar region.     Avaliação da qualidade de vida dos pacientes totalmente desdentados reabilitados com prótese tipo protocolo

    Conventional total prostheses are often the most commonly used treatment due to their low cost. However, the use of this type of prosthesis for many years, especially the lower total prosthesis, usually leads to patient dissatisfaction due to its instability during speech and chewing. Therefore, the use of osseointegrated implants for the rehabilitation of these patients becomes attractive. The objective of this study was to evaluate the rehabilitation with fixed prosthesis type protocol on three or four implants in fully edentulous mandibles using the quality of life questionnaire. Patients aged between 57 and 82 years old were included in this study. The patients were evaluated for the satisfaction of the rehabilitation with the inferior protocol and upper total prosthesis. The evaluation of patient satisfaction was performed before the installation of the prosthesis and one year after the rehabilitation of patients, it can be observed that there is a significant improvement in the general aspects in the quality of life of these patients.     Ocorrência de ansiedade em pacientes com câncer de cabeça e pescoço: uma revisão de literatura

    Patients with head and neck cancer (CCP) may present physical and emotional symptoms during and after treatment that affect their quality of life. Studies show that anxiety is one of the major emotional disorders in patients with this malignant neoplasm. Objective: To review the literature of the last 20 years on the main studies that evaluated anxiety levels in patients with head and neck cancer to evaluate the percentage of anxiety occurrence in patients with CCP and studied the association of anxiety levels with demographic, behavioral and clinical-pathological variables. Method: Search the PUBMED database of clinical trials in humans that have used standardized instruments for symptom assessment and diagnosis of anxiety in patients diagnosed with PCC. Results: A total of 50 studies were selected. Studies of this review used standardized instruments for the evaluation of anxiety in patients with CCP. The occurrence of anxiety varied from 1% to 88.1% in the different studies when patients with CCP were evaluated after diagnosis of the disease and from 11% to 87% during and after the end of cancer treatment. Studies have shown that women, patients less than 65 years of age, smokers and alcoholics had higher levels of anxiety. An association was found that anxiety levels were associated with clinical staging of the tumor and with the presence of pain at the lesion site. Conclusion: The results reveal that a great part of the patients with CCP present some level of anxiety and that the demographic, behavioral and clinical-pathological variables influence the levels of this emotional disorder

    Capacidade protetora de dentifrícios no desgaste erosivo de restaurações com resina composta e cimento de ionômero de vidro em dentina: estudo in vitro

    The aim of this in vitro study was to evaluate the stability of the different restorative materials to adjacent dentin submitted to erosive/abrasive challenge with different dentifrices. One hundred and twenty fragments was obtained the bovine incisors were cut and polished. Half of the specimens were restored with resin-modified glass-ionomer (RMGIC) and half with composite resin (RC). After the restorations, a hemiface of each sample was protected with acid-resistant varnish. The two groups were subdivided according to the dentifrice used (n=10): without fluoride (SF negative control), sodium fluoride (NaF - positive control) and stannous fluoride (SnF). The specimens were immersed in the slurry of the toothpaste for 2 min, including 15 s of simulated brushing. The erosive challenge was performed 4x / day for 2 minutes (citric acid 0.05 M, pH 3.2), with an interval of 1 hour during 5 days. After the first and last daily challenge, the specimens were subjected to the abrasive challenge. In sequence, the resistant acid varnish was removed for the Martens hardness (HMV) and elastic modulus (Eit) analyzes of the surface of the restorative material and the adjacent dentin. In addition, the surface topography of the specimens was evaluated in Scanning Electron Microscopy (n = 3). The data were analyzed for normality and submitted to 2-way ANOVA, followed by Tukey's test for multiple comparisons. The comparison between test and control sides was done with the t test. The level of significance was 5%. In relation to the HMV, for the control side, there was only statistical difference in the dentin adjacent to the RMGIC, where the SF had values lower than the NaF and the SnF was similar to the other dentifrices.     Avaliação do processo de reparo alveolar em ratas com deficiência de estrógeno tratadas com OPG-Fc: análise imunoistoquímica, por microtomografia computadorizada e microscopia confocal

    The purpose of this study is to evaluate the bone repair formed after the upper incisor extraction of ovariectomized rats treated with OPG-Fc, simulating the action of Denosumab, drug used in anti-osteoporosis therapy. Thirty six (36) adult rats were divided into three experimental groups: 12 sham rats (SHAM), 12 ovariectomized rats (OVX/ST) and 12 ovariectomized rats treated with OPG-Fc (OVX/OPG-Fc). On day 0, ovariectomy surgery or fictional surgery was performed, after 30 days the treatment with OPG-Fc was started by subcutaneous injection (10mg/kg/2 times per week). After 30 days of medication, was performed the surgery of extraction of the upper right incisor. The euthanasia occurred 60 days after the extraction surgery, in wich the jaws containing the alveoli under repair was removed in order to perform microtomographic analysis (micro-CT), for the parameters of BV/TV, Tb.Th, Tb.N, Tb.Sp, and Po(Tot); immunohistochemical analysis directed to immunomarkers: OPG, RANKL, TRAP e Osteocalcin; and confocal microscopy evaluation, to determine the NBA (neoformed bone area) and MAR (mineral apposition rate). Quantitative data from micro-CT and confocal microscopy (NBA and MAR) were submited to homoscedasticity analysis (Shapiro-Wilk test) to distinguish parametric and nonparametric data. The parametric data was compared by ONE WAY ANOVA test and Tukey post-test, and the significance level was set at p<0.05. By immunohistochemical analysis, the OPG protein showed light to moderate labeling in the SHAM group and moderate in the OVX and OVX / OPG-Fc groups. For RANKL, moderate immunostaining was observed in the SHAM group, intense in the OVX group and mild in the OVX / OPG-Fc group. For OCN, moderate labeling was observed in all groups.     A influência do gênero no efeito da oclusão traumática no periodonto

    Steroid sex hormones are believed to play an important role in the growth, maturation and turnover of the skeleton, including the alveolar bone. This study aims to evaluate the gender factor in the effects of traumatic occlusion on the periodontium. Eighty Wistar rats aged 12 weeks were divided into four groups: a male (CM, n=20) and female (CF, n=20) Control group, and a male (TOM, n=20) and female (TOF, n=20) Traumatic Occlusion group. TO was created with a direct composite resin filling and metallic wire on the occlusion surface of the Right Mandibular First Molar. Specimens were collected at 7 and 30 postoperative days, histological sections were stained by HE and picrosirius methods, followed by histological evaluation in light or polarization microscopy, and statistical analysis using the Mann Whitney test, considering p> 0.05 as significant. TOF group show significative changes in the decrease of bone area at 7 days and 30 days; and increase in the extracellular matrix at 30 days. TOM group show significative changes in the decrease of extracellular matrix area and an increase of nuclear profile at 7 days. Also show, an increase of periodontal ligament thickness and collagen fibers type I; and a decrease in the area of collagen fibers type III and bone at 7 and 30 days. Traumatic occlusion caused bone degradation in female and male rats; however changes such as an increase of periodontal ligament thickness, increase of nuclear profile and collagen fibers type I; and a decrease in the area of collagen fibers type III of the periodontal ligament were expressive only in the male gender

    Influência da quimioterapia na expressão de citocinas pró-inflamatórias tecidual na periodontite experimental

    This study comparatively evaluated the influence of the chemotherapeutic agent Cisplatin (CIS), both over the healthy periodontium and the progression of experimental periodontitis (EP), as well as kidney functions of the rats with EP under chemotherapy. Materials and Methods: 120 male rats were distributed in 4 groups (n=30). NEP-SS group: animals that received injections of 0,5ml of 0,9% saline solution (SS) without EP induction (Sham group). EP-SS group: animals that received injections of 0,5 ml of SS and induction of EP after de first injection. NEP-CIS group: animals that received CIS injections without EP induction. EP-CIS group: animals that received CIS injections and PE induction after the first injection. For EP induction a cotton thread number 24 was fitted around the first left lower first molar. After 07, 15 and 30 days after the first intraperitoneal injection (SS or CIS), animals were euthanized. The collected mandibles were submitted to immunohistochemical analysis in the furcation region of the first lower molar for evaluation of the biomarkers TNFα and IL1β. Blood samples were collected for biochemical analyzes of creatinine and urea, before injections and at 07 and 30 days. Data were submitted to statistical analysis (p<0,05). Results: EP-CIS showed greater intensity and extension of the inflammatory response, and decreased repair process compared with group EP at all experimental periods. EP-CIS had significantly increased levels of urea and showed no significant difference in creatinine' levels. Conclusion: CIS influenced the increased expression of proinflammatory cytokines in healthy periodontal tissues, as well ass, in presence of EP, exacerbated the inflammatory process in the furcation region. Additionally, EP increased serum levels of urea, indicating an intensification of the renal damages caused by CIS

    Avaliação da associação beta tricálcio fosfato e BMP2 no reparo ósseo de alvéolos dentais curetados

    The presente study have the aim to evaluate the osteoinductive and osteoconductive effects of the beta tricalcium phosphate (TCP) and bone morphogenetic protein (rhBMP-2) association used to fill the upper incisor extraction socket in order to analyse cellular responses in the presence of absence of periodontal ligament remanants of the alveolar wall. It was used 24 rats, divided in two groups: with periodontal ligament or with no periodontal ligament. Each group was divided in two sub groups, according to the material to be studied into the dental socket (Bone Ceramic alone or in association with BMP-2). After general anestesia induction through intramuscular infiltration, animals had right upper incisor extracted, with an instrument that was specially adapted and after the socket filling with the different materials, it was performed the suture with 910 polyglactin thread. The sub-groups were: Bone Ceramic with Periodontal Ligament (LP-BC), Bone Ceramic without Periodontal Ligament (SLP-BC), Bone Ceramic BMP with Periodontal Ligament (LP-BC+BMP), Bone Ceramic BMP without Periodontal Ligament (SLP-BC+BMP) . To each experimental sub group, the animals were distributed in order to evaluate the periods of 28 days after tooth extraction: 6 animals per period. After the experimental periods that were determined, the animals were sacrified, in order to obtain the samples that were fixed in 4% formaldehyde, through 48 hours. In sequence, they were washed in running water through 24 hours and then, they were kept in 10% EDTA to the demineralization step. At the end of this period, samples passed through routine laboratorial processing, in order to perform the paraffin inclusion and after this, to obtain histological sections with 5 micrometer thickness.     Reabilitação oral com facetas estéticas: cerâmica ou resina?

    In this work, a literature review was carried out on direct facets in composite resin and indirect facets in porcelain, in order to compare the advantages and disadvantages of direct (composite resin) and indirect (ceramic) techniques to determine the indications for each type of treatment. The work was carried out through a bibliographical survey, using scientific articles published between the years 2000 and 2016, as well as books and other scientific publications. Two clinical cases are presented with the purpose of exemplifying and comparing these techniques and materials. It is known that direct facets of composite resin and indirect facets of ceramics have been shown to be techniques with excellent clinical results, and it is the patient that provides the main indication for the treatment, since it is so that the patient's desire and self-perception of the need for change the look of your smile that will define the treatment to be followed by the dentist

    The objective of the present study was a brief review of the Obstructive Sleep Apnea and Hypopnea Syndrome (OSAHS) in children and adolescents, and a clinical case demonstrating the importance of correcting maxillary atresia in the treatment of this syndrome and its influence on using a device for rapid maxillary expansion in a 9-year-old male patient. Through the clinical case presented and the literature, it was concluded that orthodontic treatment, with rapid expansion of the maxilla results in an increase in respiratory capacity, with improved airflow

    Raspagem e alisamento radicular associado à terapia fotodinâmica antimicrobiana: avaliação da efetividade e da segurança no tratamento da periodontite em ratos tratados com dose oncológica de zoledronato

    The aim of the present study was to evaluate the efficacy and safety of aPDT as a coadjuvant to scaling and root planning (SRP) in the treatment of experimental periodontitis (EP) in rats treated with oncological dosage of zoledronate. Sixty rats (6 months old) were divided into three experimental groups: EP-NTL, EP-SRP and EP-SRP-aPDT. The drug treatment plan lasted 8 weeks. The rats received intraperitoneal injections of 0,45 ml of 0,9% sodium chloride solution plus 100 μg / kg zoledronate at a four day interval between applications. On the 14th day of drug treatment, a ligature was placed around the lower left first molar of each animal. On the 28th day, the ligature was removed and performed: no local treatment in the EP-NTL group; only one SRP session in the EP-SRP group and; one SRP session and three aPDT sessions, in the PE-RAR-aPDT group. The aPDT sessions were performed at 0, 2 and 4 days after removal of the ligature. Euthanasia was performed 3 and 6 weeks after ligature installation, which for the EP-SRP and EP-SRP-aPDT groups corresponded to 7 and 28 days after periodontal treatment. Hemi-mandible histological processing was performed and histological sections were submitted to hematoxylin-eosin (HE) staining. Histopathological analysis of periodontal tissues, histological analysis of percentage of bone tissue (PBT) and percentage of non-vital bone (PNVB) were performed in the furcation region of the lower left first molar. The results were analyzed by light microscopy. Quantitative data analysis and statistical analysis were carried out with a significance level of 5%. The post treatment tissue repair process was more favorable in EP-SRP-aPDT. There was no statistically significant difference in PBT, however, PNVB was lower in EP-SRP-aPDT. It is concluded that the use of aPDT as adjunctive therapy to SRP has been shown to be more effective and safer for the treatment of EP in rats

    Efeito da fibrina rica em plaquetas (FRP) sobre o processo inflamatório e reparador associada ou não a um anti-inflamatório não esteroidal (AINE) na calvária de ratos

    The objective of this study was to evaluate the effect of Rich Fibrin on Platelets (FRP) in the inflammatory process in tissue repair in calvaria of rats and its consequent tissue repair associated or not with a nonsteroidal anti-inflammatory drug (NSAID). In this study, 96 adult rats (Rattus norvegicus, albinus, Wistar) were weighed between 400 and 450g. The animals were randomly divided into 4 groups (n = 6), in which the critical size defect (5 mm diameter) filled with blood clot was performed in the clot (GC) group; non-steroidal anti-inflammatory group (GC + NSAID) with critical size defect filled with blood clot and given ketoprofen (10mg / kg / day); the critical-defect platelet-rich fibrin group (FRP) filled with autologous platelet-rich fibrin preparation; and the non-steroidal anti-inflammatory platelet-rich fibrin group (CRP + NSAID) with critical size defect filled with autologous platelet-rich fibrin preparation and administered ketoprofen intramuscularly (10mg / kg / day) for 3 days post operative. Each group was evaluated at periods of 2, 7, 14 and 28 days by means of histological analyzes of neoformed bone area (AON) and count of inflammatory cells by the IMAGE J program. The histometric results showed greater bone formation and a greater modulation of the process inflammatory from the initial to the later periods for the FRP group alone when compared to its association with NSAID, thus evidencing a delay in tissue repair for the latter group. The NSAID group alone did not contain the inflammatory process even in the later periods of analysis, thus promoting a delay in bone repair. It was concluded that the FRP without association with the NSAID was favorable in the analyzes of the experimental periods, aiding in the inflammatory response and promoting a greater bone neoformation

    Instrumentos para análise da ocorrência de trauma na infância: revisão da literatura

    Studies show that childhood trauma not only predisposes the individual to psychopathologies in adulthood, but also are associated with the maintenance of long-term emotional disorders. OBJECTIVE: The purpose of this study was to verify which are the main instruments available in the scientific literature to evaluate the history of childhood trauma in adults and to carry out a brief review of the most frequently used questionnaires. METHODS: A review of the narrative literature was carried out, where inclusion criteria were studies that: 1) were published in the English language; 2) were carried out between 2008 and 2018; 3) had abstracts available for the research; 4) made the retrospective evaluation of childhood trauma in adults; 5) collected data on traumatic events through an interview or questionnaire. We excluded from the study articles that did not provide enough information about the evaluation instruments or that did not use an interview or questionnaire to assess childhood trauma. RESULTS AND DISCUSSION: Initially 4269 articles were found in the research using the keywords. The studies were evaluated and 442 articles met the inclusion criteria. Were evaluated 29 instruments that analyze the occurrence and frequency of childhood trauma and the most used in the studies were used the Childhood Trauma Questionnaire (CTQ) in 77.60%, followed by the Childhood Experience of Care and Abuse (ECSC-Q) used in 5.65 % of the studies, Adverse Childhood Experiences Questionnaire (ACE-Q) in 3.16% of the articles and finally the Early Trauma Inventory (ETI) used...

    Toxicidade celular e ação do EGCG sobre biofilme simples e misto de microrganismos de interesse endodôntico

    One of the main causes of failure in endodontic therapy is the persistence of microbial infection even after chemical-mechanical preparation and application of intracanal medication. Many authors have explored the use of herbal medicines in order to obtain new compounds that have therapeutic properties without causing microbial resistance. Among these are the flavonoids, natural phenolic compounds which have different properties such as antimicrobial, antioxidant, osteogenic and antiosteoclastogenic. One of these flavonoid that has received attention in the literature is epigallocatechin-3-gallate (EGCG), which has also shown effect against oral pathogens. Thus, the objectives of this study were to evaluate the antimicrobial activity of EGCG on planktonic culture and simple or dual-species biofilms of microorganisms resistant to endodontic treatment, as well as evaluate its toxicity on culture of fibroblasts. Tests were performed in the planktonic culture to determine the minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) of EGCG against Enterococcus faecalis, Streptococcus mutans, Actinomyces israelii, Pseudomonas aeruginosa and Fusobacterium nucleatum and also biofilm assays with some of these microorganisms in combination with E. faecalis. In addition, the cytotoxicity of EGCG was determined on L-929 fibroblast line after 24 h exposure using the MTT assay. Data were statistically analyzed using ANOVA/Tukey tests, considering p<0.05. The results showed that EGCG showed a bactericidal effect against all the evaluated species, with values of CBM varying between 0.03 and 2 mg/mL. except by P. aeruginosa. The elimination of the viable cells from the single biofilm of S. mutans and F. nucleatum and reduction of the biofilms of E. faecalis and A. israelii were observed after treatment of the biofilms with EGCG.
