Xyridaceae and Mayacaceae are related families, which are included in the Poales. Xyridaceae is sister-group of Eriocaulaceae and both families form the xyrid clade. Mayacaceae has an unstable phylogenetic position, emerging either in the xyrid clade, or in the cyperid clade. This study aimed to analyze the embryogenesis and post-seminal development of Xyridaceae and Mayacaceae in a comparative approach to better understand the interfamilial relationships and the evolution of some embryo and seedling traits within Poales. The floral development and anatomy were also analyzed in Xyris to elucidate the evolution of gynoecium in the genus, which is divided into three sections based on ovary traits such as number of locules and type of placentation. Embryo development of the Asterad-type, differentiated embryo and formation of suspensor are ancestral conditions in Poales. Xyridaceae and Eriocaulaceae share the embryogenesis of Asterad-type, whereas the embryogenesis of Onagrad-type links Mayacaceae to the cyperid families. In addition, our results confirm that the seeds of Mayacaceae are non-operculate, a trait that has been questioned and pointed out as a probable synapomorphy of the xyrid clade. Compact cotyledon is the ancestral condition and a common character state within Poales, from which blade-like hypophyll and coleoptile have derived in Xyris and in Mayacaceae, respectively. Coleoptile is another feature shared by Mayacaceae and Cyperaceae, and it may also support the positioning of Mayacaceae in the cyperid clade. Regarding the gynoecium in Xyridaceae, axile placentation is the most likely state at the base of the family, from which intrusiveparietal placentation has derived in the Abolbodoideae. The axile placentation is the ancestral condition in Xyris and shift to parietal placentation in Xyris sect. Xyris, and to free-central or basal placentation in Xyris sect. Nematopus by shortening and sterilization of the synascidiate zone and elongation of the fertile symplicate zone during the evolution of the genus.
