Toxicidade e riscos ecológicos das tintas anti-incrustantes de terceira geração: Toxicidade e efeitos da diclofluanida sobre organismos marinhos

Resumo

Centenas de compostos são detectados nos diferentes compartimentos ambientais podendo ser de origem natural ou antrópica. Em muitos casos, pouco se sabe a respeito desses compostos, não havendo regulações sobre seu uso, e por isso são denominados compostos de interesse emergente. Dentre eles podem ser citados os biocidas das tintas anti-incrustantes. Tais tintas são utilizadas para proteger e combater a formação e o estabelecimento de comunidades bioincrustantes sobre superfícies expostas à água do mar. Elas são aplicadas em diversas estruturas, tais como embarcações, plataformas petrolíferas, tubulações submarinas, comportas de represas, tanques destinados à aquicultura, entre outras. Assim, essas tintas são utilizadas com o intuito de preservar as estruturas, bem como, a manter a navegabilidade, no caso das embarcações. O composto de interesse para este estudo é um dos biocidas utilizados nas tintas anti-incrustantes, a diclofluanida. Devido as suas características físico-químicas, parte da diclofluanida liberada na coluna d’água tenderá a adsorver ao material particulado e se depositará nas camadas sedimentares. Desta forma este estudo teve o objetivo de avaliar a toxicidade e efeitos subcrônicos da diclofluanida em exposições realizadas em água e sedimento com espécies marinhas. Além disso, experimentos utilizando sedimento também tiveram o objetivo de comparar a influência de diferentes concentrações de matéria orgânica sobre a toxicidade de sedimentos contendo diclofluanida. Para os testes ecotoxicológicos em água foram utilizados embriões do ouriço-do-mar da espécie Echinometra lucunter e fêmeas ovígeras do copépodo Nitocra sp. Já os testes em sedimento foram conduzidos com o copepódo Nitocra sp. e com o anfípodo Tiburonella viscana. No teste de exposição subcrônica à soluções aquosas foram utilizados bivalves da espécie Perna perna, e para sedimentos fortificados foram usados bivalves adultos de Anomalocardia flexuosa. Em seguida, foi realizada a técnica da retenção do vermelho neutro (apenas para P. perna), sendo também analisados o dano em DNA, teores de LPO, GSH, e atividades de GST, EROD, AChE e GPx para ambas as espécies. Para os sedimentos também foram realizadas as análises granulométricas, teor de carbonato de cálcio e de matéria orgânica. Nos testes ecotoxicológicos com soluções aquosas, os níveis tóxicos foram superiores às concentrações ambientais. Embriões de E. lucunter apresentaram maiores porcentagens de larvas anormais a partir da concentração de 1 μg/l, que foi a menor concentração de efeito observado (CEO); a Concentração Efeito Não Observado (CENO) foi de 0,1 μg/l. A CE50–40h foi de 198,5 μg/l e CE10–40h foi de 0,3577 μg/l. Nos testes com Nitocra sp., a CEO foi de 100 μg/l e a CENO foi de 10 μg/l. A CE10-7d foi de 0,0635 μg/l enquanto a CE50-7d foi de 583,83 μg/l. Nos testes de sedimento, as concentrações testadas não foram tóxicas ao copépodo Nitocra sp., e as fecundidades foram semelhantes quando comparados os dois tipos de sedimentos. Para T. viscana as taxas de mortalidade aumentaram 40% na concentração de 1000 ng/g e 30% na concentração de 10000 ng/g. Ao comparar os dois sedimentos, a maior diferença entre as mortalidades foi observada em 1000 ng/g, sugerindo que há uma influência da matéria orgânica na toxicidade da Diclofluanida. Essas investigações mostram que a diclofluanida mostra-se tóxica aos invertebrados marinhos e que, nos sedimentos, sua toxicidade pode ser influenciada pelo nível de matéria orgânica. Para as exposições subcrônicas em água, foi possível observar efeito nas maiores concentrações no vermelho neutro denotando perda da estabilidade lisossômica. Para os biomarcadores também foi possível observar alterações nas glândulas digestivas e brânquias. Para sedimento é possível prever efeito em ambos os órgãos internos e há um incremento dos efeitos nos sedimentos com maiores teores de matéria orgânica. No presente estudo foi possível constatar que o diclofluanida foi responsável por gerar respostas em todos os ensaios de toxicidade realizados em água e apenas um efeito agudo no sedimento com alto teor de matéria orgânica. Os dados de biomarcadores corroboram com os testes de toxicidade. Em água é possível observar um efeito aos organismos nas maiores concentrações testadas e em sedimento é possível observar um aumento de efeito nos organismos nos sedimentos ricos em matéria orgânica.
Hundreds of compounds have been detected in different environmental compartments, which may be natural or anthropogenic origin. In many cases, little is known about these compounds, thus there is not regulations for them, leading them be considered effective contaminants and so are called emerging. Among these compounds can be cited the biocides of antifouling paints. They are used to protect and combat the formation and establishment of biofouling communities on surfaces exposed to seawater. They are applied in various structures, such as vessels, oil platforms, submarine pipelines, tanks for aquaculture, among others. Thus, these paints are used with the intention of preserving the structures, as well as maintaining navigability in the case of vessels. The compound of interest for this study is one of the biocides used in antifouling paints, dichlofluanid. Due to its physico-chemical characteristics, part of the dichlofluanid released in the water column tended to adsorb to the particulate material and then deposit in the sedimentary layers. In this way, this study aimed to evaluate the toxicity and subchronic effects of dichlofluanid exposures, after in water and sediment with marine species. In addition, sediment experiments also aimed to evaluate how different concentrations of organic matter in the sediments could influence on their responses. Embryos of the sea urchin Echinometra lucunter and ovigerous females of the copepod Nitocra sp for the ecotoxicological tests in water. The sediment tests were conducted with the copepod Nitocra sp. and with the amphipod Tiburonella viscana. For the exposure tests in water were used bivalves of the species Perna perna and for sediment with bilvalves of Anomalocardia flexuosa. Then, the neutral red retention technique was applied only to P. perna and DNA damage, LPO, GSH, GST, EROD, AChE and GPx for both species. For the sediments were also carried out the granulometric analyzes, calculation of the content of carbonates and organic matter. In ecotoxicological tests with aqueous solutions, the toxic levels were higher than the environmental concentrations. Embryos of E. lucunter presented higher percentages of abnormal larvae from the concentration of 1 μg/l, which was the lowest observed effect concentration (LOEC); the No Observed Effect Concentration (NOEC) was 0.1 μg/l. EC50-40h was 198.5 μg/l and EC10-40h was 0.3577 μg/l. In the Nitocra sp. Tests, LOEC was 100 μg/l and NOEC was 10 μg/l. EC10-7d was 0.0635 μg/l while EC50-7d was 583.83 μg/l. In the sediment tests, the concentrations tested were not toxic to the Nitocra sp and the copepod fecundities were similar when compared the two sediment types. For T. viscana, mortality rates increased by 40% at the concentration of 1000 ng/g and 30% at the concentration of 10000 ng/g. When comparing the two sediments, the greatest difference between mortalities was observed at 1000 ng/g, suggesting that there is an influence of organic matter on the toxicity of Diclofluanid. These investigations show that diclofluanid is toxic to marine invertebrates and that, in sediments, its toxicity can be influenced by the level of organic matter. For the subchronic exposures in water, it was possible to observe effect in the higher concentrations in the neutral red denoting loss of the lysosomal stability. For the biomarkers it was also possible to observe changes in the glands and digestive gills. For sediment it is possible to predict effect in both organs as well, and there is an increase in effects on sediments with higher organic matter contents. In the present study it was possible to verify that dichlofluanid was responsible for generating responses in all toxicity tests performed in water and only an acute effect on the sediment with high organic matter content. Biomarker data corroborate with toxicity tests. In water it is possible to observe an effect to the organisms in the highest concentrations tested and in sediment it is possible to observe an increase of effect in the organisms in the sediments rich in organic matter.
