Resumo

As infecções causadas pelo vírus sincicial respiratório humano (hRSV) podem provocar sintomas leves e indistinguíveis de resfriados comuns em adultos e crianças saudáveis, mas também é uma das principais causas de infecções agudas do trato respiratório inferior, como pneumonia e bronquiolite, em recém nascidos prematuros, crianças de até dois anos e idosos. A infecção por hRSV não garante proteção contra contaminações subsequentes, e o desenvolvimento de terapias antivirais efetivas ou vacina preventiva ainda não estão disponíveis. Neste sentido, faz-se necessária a busca por alvos proteicos importantes para o combate da infecção viral e de potenciais inibidores destas proteínas. A proteína Não-Estrutural 1 (NS1) é de suma importância para o sucesso da replicação viral, capaz de reduzir a resposta imunológica, em particular a resposta do interferon do tipo I, e prevenir a apoptose celular, sendo um possível alvo de tratamento contra a infecção por hRSV. Mediante a isto, o estudo sobre a interação da NS1 com ligantes para o seu bloqueio pode possibilitar o entendimento dos mecanismos de inibição, nos quais os flavonoides têm sido descritos como sendo supressores eficazes. Além disso, essas moléculas são potenciais fármacos pelo seu baixo custo de obtenção e poucos efeitos adversos. O objetivo deste estudo foi investigar a interação entre a proteína NS1 do hRSV com alguns flavonoides utilizando espectroscopia de fluorescência e docking molecular. A NS1 foi expressa, purificada e quantificada por espectroscopia de UV/VIS e dicroísmo circular. A flexibilidade estrutural foi verificada por dinâmica molecular. A análise da interação proteína/ligante foi realizada por espectroscopia de fluorescência. A interação da NS1 com os flavonoides estudados teve maior contribuição devido a interações hidrofóbicas. Os resultados sugerem que a interação depende das estruturas dos flavonoides e da posição de suas hidroxilas. Entretanto, a ordem de grandeza de ligação foi moderada (104 M-1) para todas as moléculas estudadas, com isso o flavonoide poderia interagir com a NS1 impedindo, momentaneamente, a proteína de bloquear as vias do IFN-α, e logo desassociarem para que o flavonoide possa ser complexado por outras proteínas NS1.
Human respiratory syncytial virus (hRSV) infections can cause mild symptoms of common colds in adults and children, but it is also a leading cause of acute lower respiratory tract infections such as pneumonia and bronchiolitis in premature newborns, children until two years old and elderly people. The hRSV infections does not guarantee protection against subsequent contamination, and the development of effective antiviral therapies or preventive vaccine is not available yet. Therefore, it is necessary to search for important proteins targets to combat viral infection and potential inhibitors of these proteins. Non-Structural 1 protein (NS1) is of utmost importance for the viral replication success, capable of reducing the immune response, in particular the interferon type I response, and preventing cell apoptosis, being a possible target of treatment against hRSV infection. It is important to study the interaction of NS1 with ligands for the blockade of this protein, and thus to use it as a possible target for treatment against hRSV infection. The study on the interaction of NS1 with ligands is important to understanding the mechanisms of inhibition, which flavonoids have been described as affective suppressors. These molecules are potential drugs because of their low cost of procurement and few adverse. The aim of this study was to investigate the interaction between the hRSV NS1 protein and some flavonoids using fluorescence spectroscopy and molecular docking. NS1 protein was expressed, purified and quantified by UV/VIS spectroscopy and circular dichroism. Structural flexibility was verified by molecular dynamics. Protein-ligands interaction analysis was performed by fluorescence spectroscopy. The interaction of the protein with the studied flavonoids had their greatest contribution due to hydrophobic interactions. The results suggest the interaction depends on the flavonoid structures, the position of their hydroxyls, and the affinity to NS1. However, the order of magnitude of binding was moderate (104 M-1) for the molecules in this study, so the flavonoid could interact with NS1 momentarily impossibility the protein from blocking IFN-α pathways, and then disassociating to that the flavonoid can be complexed by other NS1.
