Efeito do pH e da composição lipídica na interação do peptídeo antimicrobiano MP1 e seu análogo H-MP1 com membranas modelo

Resumo

Peptídeos líticos com propriedades antimicrobianas são catiônicos e atuam na membrana celular desestabilizando o empacotamento lipídico levando a célula à lise. Polybia-MP1 ou MP1 (IDWKKLLDAAKQIL-NH2) é um potente bactericida, não hemolítico e não citotóxico que também inibe a proliferação de células de câncer. Ambas as atividades, bactericida e inibitória, envolvem lipídios aniônicos e pH do ambiente diferente do fisiológico. O potencial eletrostático e o pH da solução podem modular a carga do peptídeo com presença simultânea de resíduos ácidos e básicos. Neste trabalho investigamos o efeito da modulação de carga na afinidade e na eficiência lítica em vesículas lipídicas. Para esta tarefa a eficiência do MP1 em vesículas lipídicas foi comparada com a do análogo H-MP1 em que as histidinas substituíram as lisinas. Dicroísmo circular, fluorimetria, potencial zeta e microscopia óptica foram usados para investigar a ação desses peptídeos em vesículas unilamelares grandes e gigantes (LUVs e GUVs) de diferentes composições lipídicas com o pH da solução fixado em 5,5, 6,5 e 7,4. A carga do MP1 foi menos sensível ao potencial da vesícula e ao pH da solução, enquanto a carga do H-MP1 foi fortemente modulada, sendo maior em solução ácida, mas ainda menor que MP1. A modulação da carga do H-MP1 conferiu maior afinidade e desempenho lítico em solução ácida, enquanto MP1 foi melhor em pH neutro. MP1 apresentou maior atividade de permeação nas vesículas lipídicas nos três pHs investigados em comparação ao H-MP1. O vazamento induzido em GUV única foi caracterizado por um tempo de retardo da ordem de minutos seguido pela liberação do conteúdo da GUV em alguns segundos. O tempo de retardo e a permeabilidade são dependentes do pH e da concentração de peptídeo. Os resultados confirmam que a inserção desses peptídeos na fase lipídica perturba o empacotamento de lipídios, levando à formação de poros.
Lytic peptides with antimicrobial properties are cationic and act on the lipid cell membrane, destabilizing the lipid-packing leading to the cell lysis. Polybia-MP1 or MP1 (IDWKKLLDAAKQIL-NH2) is a potent bactericide, non-hemolytic, and non-cytotoxic that inhibits cancer cell proliferation. Both the bactericide and the inhibitory activities involve anionic lipids and milieu pH different from the physiological one. The electrostatic potential and the solution pH can potentially modulate the net charge of a peptide with the simultaneous presence of acidic and basic residues. This thesis investigated the effect of this charge modulation on the affinity and on the lytic efficiency in anionic vesicles. For this task, the affinity to and leakage efficiency in anionic vesicles of MP1 was compared with those of its analog H-MP1 in which histidines substituted the lysines. Circular dichroism, fluorimetry, zeta potential, and optical microscopes were used to investigate the action of these peptides in anionic large and giant unilamellar vesicles (LUVs and GUVs) of different compositions and the solution pH fixed at 5.5, 6.5 and 7.4. MP1 net charge was less sensitive to the vesicle potential and solution pH, while HMP1 charge was strongly modulated being larger at acidic solution but still smaller than MP1. The HMP1 charge modulation imparted higher affinity and lytic performance at acidic solution while MP1 was better at neutral pH. MP1 showed higher permeating activity in lipid vesicles in the three pH investigated in comparison to H-MP1. The leakage induced in single GUVs was characterized by a lag-time of an order of minutes followed by the release of GUV content in a few seconds. The lag-time and permeability were dependent on the pH and peptide concentration. The results support that the insertion of these peptides into the lipid phase disturbs the lipid-packing leading to pore formation.
