Composição genética da raia viola Pseudobatos percellens (elasmobranchii: rhinobatidae) utilizando SNPs

A biodiversidade marinha está exposta à uma ameaça crescente diante de inúmeras transformações ambientais. Compreender como os organismos respondem a tais mudanças é uma grande preocupação para os cientistas, principalmente envolvendo estudos de diversidade genética que podem ser melhor respondidos com o desenvolvimento recente de tecnologias em genômica. O sequenciamento de nova geração transformou o campo da genômica funcional, permitindo identificar polimorfismos de nucleotídeos únicos (SNPs) em todo o genoma. Um modelo interessante de aplicação desta metodologia é representado pela espécie de raia marinha Pseudobatos percellens, popularmente conhecida como raia viola, uma vez que a falta de estudos envolvendo as populações desta espécie abordando principalmente questões de estruturação e diversidade genética. Este trabalho visou caracterizar geneticamente as populações de P. percellens que ocorrem na costa brasileira, em investigações que envolveram a identificação de possível ocorrência de estruturação genética na espécie utilizando marcadores moleculares do tipo SNPs, além de identificar conexões, diferenças e similaridades entre as populações a partir da aplicação de análises estatísticas utilizadas em genética de populações. Foi utilizadas 52 amostras de P. percellens obtidas em localidades da região litorânea dos estados do Amapá, Pará, Pernambuco, São Paulo e Paraná. A filtragem dos SNPs resultou em um conjunto final de 3.329 marcadores deste tipo. Em todas as localidades a heterozigosidade observada (Ho) apresentou um valor superior ao da heterozigosidade esperada (He) e os valores para o índice de fixação ou coeficiente de endogamia (FIS) foram negativos para todas as localidades amostradas, indicando não haver endogamia nesses grupos, confirmando um excesso de heterozigotos. Os resultados indicam uma estrutura populacional com Fst significativo e as estatísticas F (estimativas ΦST) geradas pela AMOVA revelaram a existência de heterogeneidade genética significativa. As análises Bayesiana e de DAPC revelaram pelo menos dois agrupamentos genéticos distintos de P. percellens. As taxas de migração calculadas com base nas estimativas de número de migrantes, detectou alta conectividade entre as regiões Norte (Amapá e Pará) e Nordeste (Pernambuco) pela existência de fluxo gênico entre os indivíduos dessas regiões. Da mesma forma, foi observada uma alta conectividade entre as regiões Sudeste (São Paulo) e Sul (Paraná). Entretanto, os valores de fluxo gênico entre esses dois grupos (Norte/Nordeste vs Sudeste/Sul) foram baixos. Considera-se, pois, que o comportamento de mobilidade de P. percellens poderia ter influencia nos padrões de diversidade genética encontrados com a aplicação dos marcadores SNPs analisados. A evidência de novos estoques genéticos é crítica para reforçar as políticas de conservação neste grupo de organismos que se apresenta com um grande número de espécies ameaçadas.
Marine biodiversity is a growing threat in the face of countless environmental changes. Understanding how organisms respond to such changes is a major concern for scientists, especially involving studies of genetic diversity that can be better elucidated to with the recent development of technologies in genomics. Next-generation sequencing has transformed the field of functional genomics, enabling the identification of single nucleotide polymorphisms (SNPs) throughout the genome. An interesting model for the application of this methodology is represented by the guitarfish species Pseudobatos percellens, popularly known as “guitarfish”, since the lack of studies involving populations of this species addressing mainly issues of structuring and genetic diversity. This work aimed to genetically characterize the populations of P. percellens that occur on the Brazilian coast, in investigations that involved the identification of possible occurrence of genetic structuring in the is species using molecular markers such as SNPs, in addition to identifying connections, differences and similarities between populations with the application of statistical analyzes used in population genetics. Fifty-two samples of P. percellens obtained from localities in the coastal region of the states of Amapá, Pará, Pernambuco, São Paulo and Paraná were used. Filtering the SNPs resulted in a final set of 3,329 markers of this type. For all localities the observed heterozygosity (Ho) had a higher value than the expected heterozygosity (He) and the values for the fixation index or coefficient of inbreeding (Fis) were negative for all sampled localities, indicating that there was no inbreeding in these groups and an excess of heterozygotes. The results indicate a population structure with significant Fst and the F statistics (ΦST estimates) generated by AMOVA revealed the existence of significant genetic heterogeneity, with a partition of genetic variation. Bayesian and DAPC analysis revealed at least two distinct genetic groupings of P. percellens. The migration rates calculated through migrant’s number estimated returned high connectivity between North (Amapá e Pará) and Northeast regions by the existence of gene flow among the individuals of these regions. In the same way, high connectivity between Southeast (São Paulo) e South (Paraná) areas was observed. However, the values of genetic flow between these two groups (Norte/Nordeste vs. Sudeste/Sul) were low. Therefore, it is considered that the mobility behavior of P. percellens could be an influential factor in the genetic diversity found with the application of the analyzed SNPs markers. Evidence of new genetic stocks is critical to strengthen conservation policies in this group of organisms that has a large number of threatened species.
