Estudo biológico, molecular e morfométrico das cepas Tsh 8 e Tv de Trypanosoma cruzi (Chagas, 1909) (Kinetoplastida, Trypanosomatidae)

Abstract

A doença de Chagas é uma zoonose que atinge mais de 7 milhões de pessoas. Trypanosoma cruzi, agente etiológico dessa doença, é um protozoário flagelado, com ciclo evolutivo que compreende hospedeiros mamíferos e insetos hematófagos, os triatomíneos. Este projeto teve como objetivo o estudo biológico, molecular, e morfométrico das cepas Tsh 8 e Tv de Trypanosoma cruzi. O estudo molecular das cepas Tsh 8 e Tv foi realizado pela técnica de FFLB, que indicou que pertencem à mesma DTU I. O estudo biológico da cepa Tv de T. cruzi consistiu na determinação do período pré-patente, pico parasitêmico, taxa de mortalidade, período de desaparecimento das formas circulantes no sangue periférico e período de sobrevida de camundongos Swiss infectados. O estudo morfométrico das formas epimastigotas da cepa Tsh 8 e das formas tripomastigotas e epimastigotas da cepa Tv foi feito a partir da mensuração dos parâmetros: Comprimento total do parasito (CT), comprimento da célula (CC), flagelo livre (FL), largura da célula (LC), área do cinetoplasto (AC), área do núcleo (AN), distância da extremidade anterior ao núcleo (NA), distância da extremidade posterior ao núcleo (PN) e índice nuclear (IN=PN/NA).

Chagas disease is a zoonosis that affects more than 7 million people. Trypanosoma cruzi, the etiological agent of this disease, is a flagellated protozoan with a life cycle that involves mammalian hosts and hematophagous insects, the triatomines. This project aimed to study the biological, molecular, and morphometric characteristics of the Trypanosoma cruzi strains Tsh 8 and Tv. The molecular analysis of the Tsh 8 and Tv strains was performed using the FFLB technique, which indicated that they belong to the same DTU I. The biological study of the Tv strain of T. cruzi involved determining the pre-patent period, parasitemia peak, mortality rate, disappearance period of circulating forms in peripheral blood, and the survival period of infected Swiss mice. The morphometric study of the epimastigote forms of the Tsh 8 strain and the trypomastigote and epimastigote forms of the Tv strain was carried out by measuring the following parameters: total length of the parasite (CT), cell length (CC), free flagellum (FL), cell width (LC), kinetoplast area (AC), nucleus area (AN), distance from the anterior end to the nucleus (NA), distance from the posterior end to the nucleus (PN), and nuclear index (IN = PN/NA).
