O uso intensivo de herbicidas em sistemas agrícolas tropicais tem ampliado as preocupações quanto aos riscos ecotoxicológicos associados à exposição de organismos não-alvo em compartimentos terrestres e aquáticos. O objetivo geral deste estudo foi investigar os efeitos ecotoxicológicos dos herbicidas indaziflam e isoxaflutol, aplicados como formulações isoladas e mistura em organismos terrestres e aquáticos, a fim de delimitar as implicações ambientais associados ao uso desses produtos. Foram estabelecidos dois objetivos específicos: (i) avaliar os efeitos dos herbicidas indaziflam e isoxaflutol, aplicados como formulações isoladas e mistura sobre organismos edáficos não-alvo (Proisotoma minuta e Enchytraeus crypticus) utilizando um solo natural (latossolo vermelho) e o solo artificial tropical; e (ii) investigar a toxicidade aguda e crônica dos herbicidas indaziflam e isoxaflutol, aplicados como formulações isoladas para organismos aquáticos, por meio de testes com Daphnia magna, Ceriodaphnia silvestrii, Strandesia rondoniensis e Lemna minor, sendo os testes crônicos conduzidos exclusivamente com C. silvestrii. Os testes foram conduzidos em delineamento inteiramente casualizado, com exposição a múltiplas concentrações dos herbicidas, em condições controladas de laboratório e seguindo protocolos ecotoxicológicos padronizados para cada organismo-teste. A abordagem experimental permitiu comparar respostas ecotoxicológicas entre compartimentos terrestre e aquático, considerando diferentes níveis de organização biológica e tempos de exposição. O teste com a espécie nativa de colêmbolo, P. minuta foi conduzido por 28 dias, enquanto o teste com o enquitreídeo E. crypticus teve duração de 21 dias, para avaliação de sobrevivência e reprodução. No compartimento aquático abrangeram testes agudos (48-96 h) e crônicos, incluindo teste crônico de oito dias com o cladócero C. silvestrii, no qual foram avaliados os parâmetros sobrevivência e reprodução, além do teste com a macrófita L. minor, no qual foi avaliada a inibição do crescimento, conforme protocolos normativos específicos para cada organismo-teste. Para P. minuta, os herbicidas isolados induziram respostas bifásicas, com estímulo populacional em baixas concentrações e efeitos adversos apenas em níveis mais elevados. As concentrações efetivas, CE10 variaram de 473,3 a 678,7 mg i.a. kg-1 para o indaziflam e de 1094,9 a 1514,4 mg i.a. kg-1 para o isoxaflutol. Para o isoxaflutol, a CE50 foi estimada em 2048,6 e 2157,4 mg i.a. kg-1, enquanto a CE50 não pôde ser estimada para o indaziflam. Em contraste, a mistura apresentou CE10 entre 274,9 e 314,1 mg i.a. kg-1, CE20 entre 334,2 e 370,1 mg i.a. kg-1 e CE50 entre 466,6 e 489,9 mg i.a. kg-1, com interação sinérgica em ambos os solos. Para E. crypticus, os efeitos foram dependentes do solo e do parâmetro analisado, sendo observada, para a mistura, redução reprodutiva no solo artificial com CE10 de 456,3 mg i.a. kg-1, CE20 de 623,2 mg i.a. kg-1 e CE50 de 1728,2 mg i.a. kg-1, bem como sinergismo no solo natural. No compartimento aquático, os testes revelaram elevado contraste entre a resposta de produtores primários e de consumidores. Nos testes agudos, observou-se um gradiente consistente de sensibilidade, com L. minor como o organismo mais sensível, apresentando CE50 de 0,18 mg i.a. L-1 para o indaziflam e 0,72 mg i.a. L-1 para o isoxaflutol. Para os microcrustáceos, as CE50 do indaziflam variaram de 1,81 a 34,51 mg i.a. L-1, enquanto para o isoxaflutol os valores foram superiores a 5 mg i.a. L-1. A mistura de indaziflam + isoxaflutol apresentou toxicidade intermediária nos testes agudos, mantendo L. minor como o organismo mais sensível (CE50 = 0,92 mg i.a. L-1). Nos testes crônicos com C. silvestrii, a reprodução foi reduzida em até 94% para o indaziflam, 75% para o isoxaflutol e 93% para a mistura, a qual apresentou efeito sinérgico. Os resultados evidenciam que a resposta ecotoxicológica dos herbicidas indaziflam e isoxaflutol variou em função do organismo-teste, do compartimento ambiental e do tempo de exposição, demonstrando padrões distintos de sensibilidade entre organismos terrestres e aquáticos. Para os organismos edáficos, a mistura apresentou maior toxicidade que os herbicidas isolados. Em contraste, no compartimento aquático, os herbicidas isolados apresentaram maior toxicidade nos testes agudos, enquanto a mistura promoveu os maiores efeitos nos testes crônicos, particularmente sobre os parâmetros reprodutivos. A ocorrência consistente de interações sinérgicas e os impactos expressivos sobre a reprodução reforçam a importância de incorporar dados quantitativos de toxicidade crônica e de misturas reais de uso agrícola em avaliações de risco ambiental, especialmente em ecossistemas tropicais, nos quais a extrapolação de dados de regiões temperadas pode subestimar riscos ecológicos relevantes.

The intensive use of herbicides in tropical agricultural systems has increased concerns about the ecotoxicological risks associated with the exposure of non-target organisms in terrestrial and aquatic compartments. The overall objective of this study was to investigate the ecotoxicological effects of the herbicides indaziflam and isoxaflutol, applied as isolated formulations and mixtures, on terrestrial and aquatic organisms, in order to delineate the environmental implications associated with the use of these products. Two specific objectives were established: (i) to evaluate the effects of the herbicides indaziflam and isoxaflutol, applied as isolated formulations and mixtures, on non-target edaphic organisms (Proisotoma minuta and Enchytraeus crypticus) using a natural soil (red latosol) and a tropical artificial soil; (ii) to investigate the acute and chronic toxicity of the herbicides indaziflam and isoxaflutol, applied as isolated formulations to aquatic organisms, through tests with Daphnia magna, Ceriodaphnia silvestrii, Strandesia rondoniensis, and Lemna minor, with chronic tests conducted exclusively with C. silvestrii. The tests were conducted in a completely randomized design, with exposure to multiple concentrations of the herbicides, under controlled laboratory conditions and following standardized ecotoxicological protocols for each test organism. The experimental approach allowed for the comparison of ecotoxicological responses between terrestrial and aquatic compartments, considering different levels of biological organization and exposure times. The test with the native springtail species, P. minuta, was conducted for 28 days, while the test with the enchytraeid beetle E. crypticus lasted 21 days, to evaluate survival and reproduction. In the aquatic compartment, acute (48-96 h) and chronic tests were conducted, including an eight-day chronic test with the cladoceran C. silvestrii, in which survival and reproduction parameters were evaluated, as well as a test with the macrophyte L. minor, in which growth inhibition was evaluated, according to specific normative protocols for each test organism. For P. minuta, the isolated herbicides induced biphasic responses, with population stimulation at low concentrations and adverse effects only at higher levels. The effective concentrations, EC10, ranged from 473.3 to 678.7 mg a.i. kg-1 for indaziflam and from 1094.9 to 1514.4 mg a.i. kg-1 for isoxaflutol. For isoxaflutol, the EC50 was estimated at 2048.6 and 2157.4 mg a.i. kg-1, while the EC50 could not be estimated for indaziflam. In contrast, the mixture showed EC10 between 274.9 and 314.1 mg a.i. kg-1, EC20 between 334.2 and 370.1 mg a.i. kg-1 and EC50 between 466.6 and 489.9 mg a.i. kg-1, with synergistic interaction in both soils. For E. crypticus, the effects were dependent on the soil and the parameter analyzed. For the mixture, a reduction in reproduction was observed in the artificial soil with EC10 of 456.3 mg a.i. kg-1, EC20 of 623.2 mg a.i. kg-1, and EC50 of 1728.2 mg a.i. kg-1, as well as synergism in the natural soil. In the aquatic compartment, the tests revealed a high contrast between the response of primary producers and consumers. In the acute tests, a consistent sensitivity gradient was observed, with L. minor as the most sensitive organism, presenting EC50 of 0.18 mg a.i. L-1 for indaziflam and 0.72 mg a.i. L-1 for isoxaflutol. For microcrustaceans, the EC50 values of indaziflam ranged from 1.81 to 34.51 mg a.i. L-1, while for isoxaflutol the values were greater than 5 mg a.i. L-1. The mixture of indaziflam and isoxaflutol showed intermediate toxicity in acute tests, with L. minor remaining the most sensitive organism (EC50 = 0.92 mg a.i. L-1). In chronic tests with C. silvestrii, reproduction was reduced by up to 94% for indaziflam, 75% for isoxaflutol, and 93% for the mixture, which showed a synergistic effect. The results show that the ecotoxicological response of the herbicides indaziflam and isoxaflutol varied depending on the test organism, the environmental compartment, and the exposure time, demonstrating distinct patterns of sensitivity between terrestrial and aquatic organisms. For edaphic organisms, the mixture showed greater toxicity than the herbicides alone. In contrast, in the aquatic compartment, isolated herbicides showed greater toxicity in acute tests, while the mixture promoted the greatest effects in chronic tests, particularly on reproductive parameters. The consistent occurrence of synergistic interactions and the significant impacts on reproduction reinforce the importance of incorporating quantitative data on chronic toxicity and real-world agricultural mixtures in environmental risk assessments, especially in tropical ecosystems, where extrapolation of data from temperate regions may underestimate relevant ecological risks.
