| dc.description.abstract | estrogênica. A pesquisa compreendeu etapas in vitro e in vivo. Dessa forma, foram constituídos os grupos SHAM SO CONV, SHAM SO VK2, SHAM CO CONV, SHAM CO VK2, OVX SO CONV, OVX SO VK2, OVX CO CONV e OVX CO VK2. | pt |

| dcterms.impact | Estrogen deficiency compromises bone metabolism and microarchitecture, potentially impairing peri-implant bone healing and osseointegration. The present study evaluated the peri-implant bone tissue response to additively manufactured titanium implants, either solid or featuring a hollow osseointegration chamber, functionalized with Vitamin K2, in an experimental model of estrogen deficiency. The research comprised in vitro and in vivo phases. In the in vitro phase, osteoblastic cells obtained from the femoral bone marrow of rats (n = 9) were cultured to assess cell viability, total protein content, alkaline phosphatase activity, mineralized nodule formation, and cell morphology. In the in vivo phase, 64 female Wistar rats were allocated into eight experimental groups (n = 8 per group), according to the combination of three factors: systemic condition, sham-operated control (SHAM) or ovariectomized (OVX); implant design, solid (SO) or with a hollow osseointegration chamber (CO); and surface treatment, conventional (CONV) or functionalized with Vitamin K2 (VK2). Accordingly, the following groups were established: SHAM SO CONV, SHAM SO VK2, SHAM CO CONV, SHAM CO VK2, OVX SO CONV, OVX SO VK2, OVX CO CONV, and OVX CO VK2. The implants were placed in the tibiae, with evaluations performed at 14 and 28 postoperative days using reverse torque testing, real-time polymerase chain reaction, and microcomputed tomography. Molecular analysis included alkaline phosphatase, osteocalcin, vascular endothelial growth factor, osteoprotegerin, and receptor activator of nuclear factor kappa B ligand. Microtomographic evaluation included bone volume fraction, trabecular number, thickness, and separation, as well as intersection surface. In the in vitro phase, Vitamin K2 promoted greater cellular metabolic activity at 7 days, without a corresponding increase in osteoblastic differentiation or mineralization. In the in vivo phase, no statistically significant differences in reverse torque were observed at 14 days. Molecular analysis revealed alterations in the osteoprotegerin/receptor activator of nuclear factor kappa B ligand ratio in ovariectomized animals, whereas implants with hollow osseointegration chambers functionalized with Vitamin K2 exhibited changes in alkaline phosphatase and vascular endothelial growth factor expression. Microcomputed tomography demonstrated higher bone volume fraction and trabecular number at 28 days in control animals receiving conventional solid implants, whereas ovariectomized animals receiving functionalized hollow-chamber implants exhibited higher absolute values of trabecular thickness and number at 14 days compared with 28 days. It was concluded that estrogen deficiency altered the molecular response and peri-implant bone microarchitecture, whereas Vitamin K2 functionalization was associated with cellular and molecular changes during the early stages of bone healing, without demonstrating superior mineralization or biomechanical implant stability. | en |
