Avaliar in vitro a atividade antimicrobiana e antibiofilme do extrato glicólico
de R. centifolia L. sobre culturas planctônicas e biofilmes de Prevotella intermedia e
Parvimonas micra, a citotoxicidade e a ação anti-inflamatória em macrófagos de
camundongos (RAW 264.7) estimulados por lipopolissacarídeo (LPS) de Escherichia
coli. Material e métodos: para verificação de atividade antimicrobiana do extrato
sobre os microrganismos anaeróbios, foi aplicado teste de microdiluição, preconizado
pelo Clinical and Laboratory Standards Institute (CLSI), norma M11-A7. As
concentrações mais efetivas foram analisadas sobre biofilmes por 5 min e 24 h.
Posteriormente, foi avaliada a citotoxicidade do extrato em linhagem RAW 264.7 por
meio do ensaio colorimétrico MTT, e a ação anti-inflamatória foi verificada por ELISA,
com quantificação de citocinas pró-inflamatórias (IL-1β e TNF-α), após exposição dos
macrógafos às concentrações mais efetivas. Os resultados foram analisados pelo
teste de Kruskal-Wallis suplementado por Dunn considerando-se p≤0,05.
Resultados: o extrato de R. centifolia L. não apresentou ação antimicrobiana para P.
intermedia e P. micra. Após 24 h de tratamento, o extrato (100 mg/mL) reduziu a
viabilidade do biofilme de P. micra em 65,24%. Sobre a ação citotóxica em
macrófagos, o extrato não apresentou citotoxicidade nas concentrações 3-25 mg/mL
após 5 min e 24 h de tratamento. Com relação a ação anti-inflamatória, não houve
redução das citocinas pró-inflamatórias analisadas. Conclusão: conclui-se que o
extrato de R. centifolia L. possui ação antibiofilme contra P. micra após 24h de
tratamento. O efeito citotóxico do extrato em macrófagos foi dose-dependente e não
possui atividade anti-inflamatória.

To evaluate in vitro the antimicrobial and antibiofilm activity of the glycolic
extract of Rosa centifolia L. on planktonic cultures and biofilms of Prevotella intermedia
and Parvimonas micra, the cytotoxicity and the anti-inflammatory action in mouse
macrophages (RAW 264.7) stimulated by lipopolysaccharide (LPS) from Escherichia
coli. Material and methods: to verify the antimicrobial activity of the extract on
anaerobic microorganisms, a microdilution test, recommended by the Clinical and
Laboratory Standards Institute (CLSI), standard M11-A7, was applied. The most
effective concentrations were analyzed on biofilms for 5 min and 24 h. Subsequently,
the cytotoxicity of the extract in RAW 264.7 strain was evaluated using the MTT
colorimetric assay, and the anti-inflammatory action was verified by ELISA, with
quantification of pro-inflammatory cytokines (IL-1β and TNF-α), after exposure of the
macrophages at the most effective concentrations. The results were analyzed using
the Kruskal-Wallis test supplemented by Dunn, considering p≤0.05. Results: the
extract of R. centifolia L. showed no antimicrobial action against P. intermedia and P.
micra. After 24 h of treatment, the extract (100 mg/mL) reduced P. micra biofilm viability
by 65.24%. Regarding the cytotoxic action on macrophages, the extract did not show
cytotoxicity at concentrations of 3-25 mg/mL after 5 min and 24 h of treatment.
Regarding the anti-inflammatory action, there was no reduction in the pro-inflammatory
cytokines analyzed. Conclusion: it is concluded that the extract of R. centifolia L. has
antibiofilm action against P. micra after 24h of treatment. The cytotoxic effect of the
extract on macrophages was dose-dependent and it has no anti-inflammatory activity.
