dc.description.abstractTo evaluate the mediators of the inflammatory response, in an in vitro model, of immortalized epithelial amniotic cells (hFM-AECs) challenged by different bacterial species, alone and in combinations with Mycoplasma hominis, and the interference of this stimulus on the epithelial-mesenchymal transition. In vitro assay with hFM-AECs challenged with different species were performed in diferente moments. Supernatants were collected for quantification of IL-1β, IL-6, IL-8, IL-10, IL-13, GM-CSF, TNF-α, HBD-1, HBD-2 and HBD-3 by ELISA. For analyses of EMT, the cell shape index was applied. The data were subjected to a mixed effects model combined with parametric bootstrapping.One Way ANOVA and paired T-test were used for the EMT analysis. (a) Elevated levels of IL-6, IL-8, GM-CSF and HBD-1 were detected in relation to the reference when stimulated with Fusobacterium nucleatum, alone or in combination with M. hominis; (b) IL-13 levels were decreased in relation to the reference after stimulation with F. nucleatum, Bacteroides fragilis, Peptostreptococcus anaerobius and Streptococcus agalactiae alone and in combination with M. hominis; (c) Different isolates or combination with M. hominis stimulated EMT. F. nucleatum alone or in combination with M. hominis, have an important capacity to induce a characteristic inflammatory signature. Bacterial combinations stimulate EMT.en
