Conversão de glicose e xilose em etanol por Saccharomyces cerevisiae ATCC 26602, Zymomonas mobilis CCT 4494 e Pachysolen tannophilus CCT 1891

Resumo

O etanol pode ser obtido pela conversão de açúcares como a glicose e a xilose por diferentes micro-organismos. Empregar dois ou mais micro-organismos que assimilem estes açúcares e utilizar o método de imobilização celular em um determinado suporte tem se mostrado eficaz na produção de etanol comparado com a monocultura com células livres. Diante disto, o objetivo do presente trabalho foi comparar a produção de etanol entre células livres e imobilizadas em casca de soja em diferentes condições da levedura Saccharomyces cerevisiae ATCC 26602, da bactéria Zymomonas mobilis CCT 4494 e da levedura Pachysolen tannophilus CCT 1891 e verificar a cofermentação destes três microorganismos em meio de cultura contendo diferentes concentrações de glicose e xilose como simulação da produção de etanol de segunda geração. Para isto foram realizadas fermentações com os três micro-organismos, em monocultura, em meios de cultivo contendo glicose para a levedura S. cerevisiae e a bactéria Z. mobilis e em meios contendo xilose para a levedura P. tannophilus utilizando células livres e imobilizadas por ligação a superfície em casca de soja. Variou-se condições de cultivo como pH, temperatura, velocidade de agitação e concentração inicial de açúcar. Todas as fermentações ocorreram em 15 horas e foram realizadas análise para verificar crescimento celular, consumo de açúcar, produção de etanol e coeficiente de rendimento. Após estes experimentos, selecionaram-se as condições que proporcionaram maior coeficiente de rendimento para cada micro-organismo e foi realizado cultivo com cada micro-organismo retirando-se alíquotas a cada hora de fermentação para verificar o melhor tempo. A partir destas informações foram realizados dois tipos de cofermentações em diferentes meios de cultivo, para os quais foram variadas as concentrações de glicose e xilose. Realizou-se o reciclo celular da monocultura e da cofermentação. Para os três micro-organismos em monocultura o maior rendimento foi obtido utilizando o método de imobilização celular. Com a levedura S. cerevisiae o maior rendimento foi de 0,44 g.g-1 nas condições de 35 ºC, estático, concentração inicial de açúcar de 60 g.L-1 e pH 5,5. No reciclo desta levedura obteve-se maior rendimento (0,37 g.g-1) no ciclo 11. Utilizando a bactéria Z. mobilis foi alcançado um rendimento de 0,39 g.g-1 nas condições de 30 ºC, estático, concentração inicial de açúcar de 90 g.L-1 e pH 5,5. O reciclo promoveu uma aumento do rendimento para 0,49 g.g-1 no ciclo 5. Com a levedura P. tannophilus o maior rendimento obtido foi de 0,44 g.g-1 nas condições de 35 ºC, velocidade de agitação de 100 rpm, concentração inicial de açúcar de 60 g.L-1 e pH 4,5. No reciclo o maior rendimento foi de 0,38 g.g-1 no ciclo 5. Na cofermentação, o qual inoculou-se a levedura S. cerevisiae, seguida da bactéria Z. mobilis e da levedura P. tannophilus no meio de cultura com 40% de xilose e 60% de glicose, foi obtido o maior rendimento (0,19 g.g-1) e no reciclo da cofermentação ocorreu um maior rendimento de 0,31 g.g-1 no ciclo 3. A imobilização celular proporcionou um aumento do rendimento da produção de etanol para a levedura S. cerevisiae e para a bactéria Z. mobilis a partir da glicose e, esta foi necessária para obtenção de etanol com a levedura P. tannophilus. O reciclo da cofermentação forneceu aumento no coeficiente de rendimento no terceiro ciclo.
Ethanol can be obtained by the conversion of sugars such as glucose and xylose by different microorganisms. Employing two or more microorganisms that assimilate these sugars and use the cellular immobilization method in a given support has been shown to be effective in the production of ethanol compared to free-cell monoculture. The objective of the present work was to compare the ethanol production between free and immobilized cells in soybean hull under different conditions of the yeast Saccharomyces cerevisiae ATCC 26602, bacterium Zymomonas mobilis CCT 4494 and the yeast Pachysolen tannophilus CCT 1891 and verify the co-fermentation of these three microorganisms in culture medium containing different concentrations of glucose and xylose as simulation of the production of second generation ethanol. Fermentations have been carried out with the three microorganisms, in monoculture, in culture media containing glucose for the yeast S. cerevisiae and the bacteria Z. mobilis and in media containing xylose for the yeast P. tannophilus using free cells and immobilized by surface connection in soybean hull. The experimental conditions pH, temperature, stirring speed and initial sugar concentration have been varied. All fermentations occurred in 15 hours and analyzes were performed to verify cell growth, sugar consumption, ethanol production and yield. After these experiments, the conditions that provided the highest yield for each microorganism were selected and experiments have been carried out using each microorganism, removing aliquots at each hour to verify the best time. Thus, two types of co-fermentations have been carried out in different culture media, whose concentrations of glucose and xilose have been varied. Cellular recycling of monoculture and co- fermentation has been carried out. For the three monoculture cultivation the highest yield has been obtained using the cellular immobilization method. For S. cerevisiae, the highest yield was 0.44 g.g-1 under the conditions of 35 °C, static, initial sugar concentration of 60 g.L-1 and pH 5.5. In the recycling of this yeast the higher yield (0.37 g.g-1) has been obtained in cycle 11. Using the Z. mobilis a yield of 0.39 g.g-1 has been achieved under conditions of 30 °C, static, initial sugar concentration of 90 g.L-1 and pH 5.5. The recycle promoted an increase in yield to 0.49 g.g-1 in cycle 5. For P. tannophilus the highest yield was 0.44 g.g-1 under the conditions of 35 °C, stirring speed of 100 rpm, initial sugar concentration of 60 g L-1 and pH 4.5. In the recycle the highest yield was 0.38 g.g-1 in cycle 5. In the co-fermentation, S. cerevisiae has been inoculated, followed by Z. mobilis and P. tannophilus in the culture medium combining 40% of xylose and 60% of glucose, the highest yield obtained was 0.19 g.g-1 and in the recycle of the co-fermentation the higher yield was 0.31 g.g-1 in cycle 3. The cellular immobilization provided an increase in ethanol production yield for S. cerevisiae and Z. mobilis from glucose and the cellular immobilization was necessary to obtain ethanol using P. tannophilus from xylose. The recycling of co-fermentation provided an increase in the coefficient of yield in the third cycle.
