Avaliação da angiogênese e resposta inflamatória em enxertos cutâneos submetidos à laserterapia em ratos (Rattus novergicus albinus wistar)

Resumo

As técnicas reconstrutivas têm sido amplamente utilizadas na Medicina Veterinária. Com a intenção de reduzir complicações inerentes às técnicas reconstrutivas, são utilizadas terapias adjuvantes pós-operatórias como a laserterapia de baixa intensidade (LBI). Objetivou-se definir os efeitos da laserterapia de baixa intensidade sobre a cicatrização, inflamação e vascularização de enxertos cutâneos em intervalos de tempo aplicáveis à rotina cirúrgica veterinária. Foram utilizados 40 ratos (Rattus novergicus albinus wistar), sendo cada animal submetido à enxertia cutânea autógena em região interescapular, sendo distribuídos de forma aleatória em cinco grupos (G1, G2, G3, G4 e G5) de acordo com a dose de 6 J/cm² ou 10 J/cm² a cada 3 ou 5 dias, aplicada sobre o enxerto cutâneo durante 15 dias. Avaliação macroscópica aos dias três, sete e 15 não demonstrou alteração com a aplicação de LBI sobre os enxertos. Avaliação histoquímica com picrosirius apresentou maior expressão de colágeno tipo I – vermelho em enxertos de G5 (p<0,05), enquanto não houve expressão do mesmo em G1; a expressão de colágeno tipo III – verde não foi influenciada pela LBI. A avaliação histoquímica com hematoxilina-eosina evidenciou maior número de fibroblastos em enxertos de G4 (p<0,05) e menor hemorragia em enxertos de G5 (p<0,05). Houve modulação da resposta inflamatória em enxertos cutâneos irradiados, enquanto a LBI não teve efeito sobre a angiogênese. Conclui-se que a exposição de enxertos cutâneos à dose de energia de 6 J/cm² ou 10 J/cm² a cada 5 dias beneficiaram a cicatrização e a modulação da inflamação local.
The reconstructive techniques has been widely used in Veterinary Medicine. To reduce inherent complications to the reconstructive techniques, the postoperative adjuvants therapies are used, such as low-level laser therapy (LLLT). The aim of this work is to determine the low-level laser therapy’s effects on the healing, inflammation and vascularization of the free skin grafts grafts in applicable time intervals to the veterinary surgery routine. Forty rats (Rattus novergicus albinus wistar) were used, and each one was submitted to the autogenous free skin graft on the interescapular region. They were allocated randomly in five groups (G1, G2, G3, G4 and G5) according to the 6 J/cm² or 10 J/cm² doses every 3 or 5 days for 15 days. The macroscopic evaluation on the third, seventh and fifteenth day did not show alterations of the LLLT on the grafts. The histochemical evaluation with picrosirius had more expression of the type I collagenous – red in the grafts of G5 (p<0,05), while in the G1 did not; the expression of the type III collagenous – green was not influenced by LPLT. The histochemical evaluation with hematoxylin-eosin showed more numbers of fibroblasts in the grafts of G4 (p<0,05) and less hemorrhage in grafts of G5 p<0,05). It had modulation of the inflammatory response in the irradiated cutaneous grafts, while the LPLT had no effect on the angiogenesis. Then, it observed that the exposure of the cutaneous grafts to the 6 J/cm² or 10 J/cm² doses every 5 days improved the healing and the modulation of the local inflammation.
