Resumo
O presente trabalho descreve o projeto desenvolvido durante a realização do
Estágio em Indústria obrigatório para a conclusão da graduação em Bacharelado
em Química Ambiental Tecnológica, oferecido pela Universidade Estadual
Paulista “Júlio de Mesquita Filho”, campus de Bauru, em uma empresa localizada
em Macatuba-SP. Trata-se de uma empresa do ramo alimentício que têm o
objetivo de promover saúde e bem-estar para seres humanos e nutrição animal
por meio de processos biotecnológicos. Em um dos processos realizados, ocorre
a produção de ácido glutâmico por meio de fermentação, o qual necessita de
acompanhamento em tempo real da produção dessa substância para
estabelecer a qualidade do processo. Os métodos tradicionais utilizados são
realizados por meio de um analisador bioquímico (que emprega o uso da
membrana de glutamato) ou cromatografia líquida de alta eficiência com detector
por índice de refração (HPLC-RID). O trabalho foi desenvolvido visando explorar
pontos que apresentam potencial de melhoria. Desta forma, as atividades
executadas baseiam-se no desenvolvimento de um método analítico para
análise de ácido glutâmico livre via cromatografia líquida de alta eficiência com
detector por arranjo de diodos (HPLC-DAD), com o objetivo de otimizar os
métodos utilizados, reduzindo o desgaste de equipamentos, economia na
utilização de consumíveis, otimização do tempo, além de ampliar as opções para
realização de análises simultâneas de outros processos desenvolvidos pela
empresa.
The present work describes the project developed during the completion of the Internship in Industry, mandatory for the conclusion of the Bachelor's Degree in Technological Environmental Chemistry, offered by the São Paulo State University "Júlio de Mesquita Filho", Bauru campus, in a company located in Macatuba-SP. It is a food company that aims to promote health and well-being for humans and animal nutrition through biotechnological processes. In one of the processes carried out, the production of glutamic acid occurs through fermentation, which requires real-time monitoring of the production of this substance to establish the quality of the process. The traditional methods are performed using a biochemical analyzer (which uses a glutamate membrane) or high performance liquid chromatography with a refractive index detector (HPLC RID). The work was developed to explore points that have potential for improvement. Therefore, the activities performed are based on the development of an analytical method for the analysis of free glutamic acid by high performance liquid chromatography with diode array detector (HPLC-DAD), with the objective of optimizing the methods used, reducing the equipament detrition, savings in the use of consumables, time optimization, in addition to expanding the options for carrying out simultaneous analyzes of other processes developed by the company.
