Efeito da aplicação de ácido húmico sobre o crescimento de soja e sobre o microbioma edofítico em condições de restrição hídrica

Resumo

Bactérias promotoras de crescimento vegetal (BPCV) e ácidos húmicos (AH) são opções promissoras para reduzir o uso de fertilizantes minerais, bem como, melhorar a resposta de plantas a estresses. E embora muitos estudos tenham demonstrado os efeitos de BPCV e AH separadamente, poucas informações estão disponíveis sobre as respostas das plantas à aplicação combinada desses bioestimulantes em leguminosas, como a soja. Ademais, são praticamente inexistentes estudos que abordem os efeitos que AH exerce sobre a comunidade microbiana endofítica de plantas, e como esta relação afeta o crescimento, desenvolvimento e proteção vegetal a estresses. Neste contexto, esta tese teve o objetivo de avaliar o efeito de K-humato [KH](AH comercial) sobre a soja e seu simbionte. Para isso, diferentes doses de KH foram testadas em semente de soja inoculada com Bradyrhizobium, com intuito de observar se este composto aumenta a sobrevivência do inoculante em semente e se favorece o desenvolvimento radicular e por conseguinte, a nodulação e incremento de N na planta. A concentração de 50 mg KH L-1 foi a mais promissora e foi capaz de proporcionar maiores incrementos sobre a nodulação (número e massa de nódulos), biomassa, teor de N na parte aérea e sobrevivência do inoculante na semente, em relação ao controle. Adicionalmente, foi avaliado o papel de AH de diferentes compostos orgânicos, a saber: vermicomposto e gongocomposto, sobre o crescimento da soja e a modulação de sua microbiota sob déficit hídrico. O material vegetal foi processado para a obtenção de comunidades microbianas endofíticas. Os marcadores filogenéticos de bactérias (16S rRNA) e fungos (ITS) foram amplificados e sequenciados por tecnologia Illumina. Foi observado que em ambas as condições ambientais, os AH estimularam mais o crescimento da soja que o controle. A composição bacteriana em folhas se distinguiu com aplicação de AH, dando origem a dois grupos distintos: um formado pelo tratamento controle e outro, pelos tratamentos com os AH. Na presença de estresse, a composição bacteriana se diferiu entre os três tratamentos. Não foi observado diferenças sobre a composição da comunidade fúngica perante a utilização de AH. Táxons microbianos foram diferentemente abundantes (DA) nas plantas. Sendo a fração fúngica, mais afetada por AH em raízes estressadas. As comunidades microbianas estimuladas na presença de AH parecem estar relacionadas com a promoção direta e indireta do crescimento vegetal.
Plant growth-promoting bacteria (PGPB) and humic acids (HA) are promising options to reduce the use of mineral fertilizers, as well as improve the response of plants to stress. And while many studies have demonstrated the effects of PGPB and HA separately, little information is available on plant responses to the combined application of these biostimulants to legumes such as soybeans. Furthermore, there are practically no studies that address the effects that HA exerts on the endophytic microbial community of plants, and how this relationship affects growth, development and plant protection against stress. In this context, this thesis aimed to evaluate the effect of K-humate [KH](commercial HA) on soybean and its symbiont. For this, different doses of KH were tested in soybean seed inoculated with Bradyrhizobium, in order to observe whether this compound increases the survival of the inoculant in seed and whether it favors root development and, consequently, nodulation and N increment in the plant. The concentration of 50 mg KH L-1 was the most promising and was able to provide greater increments on nodulation (number and mass of nodules), biomass, N content in the shoot and survival of the inoculant in the seed, in relation to the control. Additionally, the role of HA of different organic compounds, namely: vermicompost and millicompost, on soybean growth and the modulation of its microbiota under water deficit was evaluated. The plant material was processed to obtain endophytic microbial communities. Phylogenetic markers of bacteria (16S rRNA) and fungi (ITS) were amplified and sequenced by Illumina technology. It was observed that under both environmental conditions, HA stimulated more soybean growth than the control. The bacterial composition in leaves was distinguished with the application of HA, giving rise to two distinct groups: one formed by the control treatment and the other, by the treatments with HA. In the presence of stress, the bacterial composition differed between the three treatments. No differences were observed on the composition of the fungal community when using HA. Microbial taxa were differently abundant (DA) in plants. Being the fungal fraction, most affected by HA in stressed roots. The microbial communities stimulated in the presence of HA seem to be related to the direct and indirect promotion of plant growth.
